Livin Gene Expression In Hepatocarcinoma And Its Correlation With Bcl-2 Expression

[Introduction] It has been suggested that suppression of apoptosis may contribute to the development and progression of cancer. Western blot method was used to study Livin protein in these tissues. Immunohistochemicai SP method was used to detect the expression of Livin and Bcl-2 proteins in these tissues. We used flow cytometry to detect the level of hepatocarcinoma cell proliferation and apoptosis and tried to draw a conclusion that the expression of Livin correlated with proliferation and apoptosis.[Methods] 1. RT-PCR for Livin mRNA 2.Western-blot analysis 3.Immunohistochemisty for Livin and Bcl-2 proteins 4. Flow cytometry to detect proliferation and apoptosis.[Results] 1. The positive rate of Livin gene was 85.2% in 54 hepatocarcinoma tissues,but Livin gene expressed low levels in 27 para-cancerous tissues, its negtive rate was 0.742% (P<0.05),the expression of both variants was simultaneous basically.The RT-PCR and Western-blot methods have identical results.2. Livin gene expression and age, sex, histological subtype (P>0.05). The expression of Livin gene was closely related to the transfer of hepatocarcinoma (P<0.05) 3. The positive rate of Livin protein was 85.2% in 54 hepatocarcinoma tissues, but Livin protein expressed low levels in 27 para-cancerous tissues,its negtive rate was 0.742% (P<0.05).The ration of positive expression of Livin was 95.34%(41/43) among the Bcl-2 protein positive expression and the ration of positive expression of Livin was 45.45% among the Bcl-2 protein negative expression (P＜0.01, r=0.252).[Conclusion] 1. Livin gene is highly expressed in hepatocarcinoma tissues,but shows litle expression in para-cancerous tissues.2. There was no significant Correlation between Livin gene expression and age,sex, Histological subtype but the Expression of Livin gene was closely related to the transfer of hepatocarcinoma 3.The expression of Livin protein was positively related to the expression of Bcl-2 protein.
