Western blot method was used to study Livin protein in these tissues. Immunohistochemicai SP method was used to detect the expression of Livin and Bcl-2 proteins in these tissues. We used flow cytometry to detect the level of hepatocarcinoma cell proliferation and apoptosis and tried to draw a conclusion that the expression of Livin correlated with proliferation and apoptosis.[Methods] 1. RT-PCR for Livin mRNA 2.Western-blot analysis 3.Immunohistochemisty for Livin and Bcl-2 proteins 4. Flow cytometry to detect proliferation and apoptosis.[Results] 1. The positive rate of Livin gene was 85.2% in 54 hepatocarcinoma tissues,but Livin gene expressed low levels in 27 para-cancerous tissues, its negtive rate was 0.742% (P<0.05),the expression of both variants was simultaneous basically.The RT-PCR and Western-blot methods have identical results.2. Livin gene expression and age, sex, histological subtype (P>0.05). The expression of Livin gene was closely related to the transfer of hepatocarcinoma (P<0.05) 3. The positive rate of Livin protein was 85.2% in 54 hepatocarcinoma tissues, but Livin protein expressed low levels in 27 para-cancerous tissues,its negtive rate was 0.742% (P<0.05).The ration of positive expression of Livin was 95.34%(41/43) among the Bcl-2 protein positive expression and the ration of positive expression of Livin was 45.45% among the Bcl-2 protein negative expression (P<0.01, r=0.252).[Conclusion] 1. Livin gene is highly expressed in hepatocarcinoma tissues,but shows litle expression in para-cancerous tissues.2.
