Objective:To observe the effects and possible mechanisms of MPO for oxidizing LDL and inducing THP-1 mononuclear macrophage foaming.Methods:The different concentration of MPO(0.5,1,1.5,2,2.5u/L)with LDL incubation;The different concentration of MPO (0.5,1,1.5,2,2.5u/L)added to H2O2 with LDL incubation,using two methods to measure MDA and REM.Added different concentration aspirin(1,5,10mmol/L)to the MPO-H2O2 system with LDL incubation, using methods to measure MDA and REM.The produce of ox-LDL by MPO pathway with the THP-1 mononuclear macrophage co-culture 24h, oil red dyeing detect cells morphology;liquid chromatography-massspectrum detect the content of cholesterin.Results:1.There are no statistically significan between simple MPO of five differents groups after LDL incubation and control group in MDA and REM(P>0.05).2.There are statistically significan between MPO-H2O2 system of five differents groups after LDL incubation and control group in MDA and REM(P<0.05),showing its can oxidize LDL and the results increasing with the concentration of MPO.3.The form of MDA and REM exists no difference when the concentration of Aspirin in lmmol/L(P>0.05),But the results sharply decreased when the concentration at 5 mmol/L and 10mmol/L;contrast to the control group,there are exists the statistically significan.4.MPO-H2O2-LDL system generate ox-LDL and THP-1 mononuclearmacrophage co-culture 24h,observing the macrophage swallow massive red lipids,there no changes in normal control group.show that the experimental group CE/TC=52.7%(>50%)contrast to CE/TC=35.70%(<50%)in normal group.Conclusion:1.MPO may be through MPO catalysis H2O2 pathway mediate the oxidation of LDL inducing THP-1 mononuclear macrophage foaming. 2.Aspirin may refrain MPO catalysis H202 pathway mediate the oxidation of LDL。...
