Objective To study the direct protective effects and the mechanism of U50488H (a selectiveκ-opioid receptor agonist) on myocardial ischemia and reperfusion in ratsMethods After setting up the model of myocardial ischemia and reperfusion in rats,32 animals were randomly divided into four different groups .i e. control group, ischemia- reperfusion group (I/R), U50488H+I/R group and Nor-BNI+U50488H+I/R group (Nor-BNI, a selectiveκ-opioid receptor antagonist) respectively. The myocardial ultra structure, the microcirculation and the incidence of ventricular arrhythmias were determined in rats.Results:①The section area of blood capillary in the U50488H+I/R group was larger than that of the I/R group and Nor-BNI+U50488H+I/R group.②The injury of the myocardial ultra structure was significantly more decreased than that of the I/R group and Nor-BNI+U50488H+I/R group .③the incidence of ventricular arrhythmia and arrhythmia score were significantly lowered than that of the I/R group and Nor-BNI+U50488H+I/R group.Conclusion: Given U50488H can significantly decrease the injury of myocardial ultra structure, improve the microcirculation and decrease the incident rate of ventricular arrhythmia through stimulatingκ—opioid during myocardial ischemia and reperfusion in rats,thus it elicits the direct protective effects during myocardial ischemia and reperfusion in rats,but the effect of U50488H was abolished by Nor-BNI.
