The Expression Of NYD-SP15 Gene With Cytidine Deaminase In ARPE19 Cells

Objective:To improve NYD-SP15 expresses in ARPE19 cells and has cytidine deaminase activity.Methods:ARPE19 cells were cultured for mRNA and proteins extraction,to improve NYD-SP 15 expresses in ARPE19 cells by RT-PCR and western blotting.Cytidine deaminase activity testing in vitro results show that NYD-SP15 has cytidine deaminase activity.Results:NYD-SP15 expresses in ARPE19 cells and has cytidine deaminase activity to hydrolyticly deaminize cytidine,deoxycytidine,and Cytarabine,to make kinds of biological effects.Conclusions:NYD-SP15 is a new gene related with cell proliferation we screened,our results show NYD-SP15 expresses in ARPE19 cells by RT-PCR and western blotting.NYD-SP15 gene has cytidine deaminase activity,which can speed up tissue metabolism,enchance DNA synthesis,and devitalize a antitumor drug Cytarabine by deaminizing.Therefore,it may participate with nosogenesis and development of tumor and proliferation diseases in eyes,as well as drug resistance of antitumor drugs such as Cytarabine.
