Objective:Oxymatrine has distinct anti-inflammatory effect for various kinds of acute exudative inflammation, but there has not been reported that showed that oxymatrine could cure chronic periodontitis up to now. The experiment, based on the tests of animals, is supposed to study the prevention and cure effects of oxymatrine on experimental periodontitis, and the effect of oxymatrine on PDLC biology behaviour under LPS existing so as to provide the experimental basis for the prevention and cure of periodontitis.The research includes the following experiments:1. The experimental study of effect oxymatrine on prohibiting alveolar bone absorption of periodontitis.Methods: twenty-five SD rats were randomly divided into three study groups as follow: the normal group, the control group and the oxymatrine administration group. Firstly, Steel-wire was placed around one of the first lower molar of the rats to induce periodontal diseases. Secondly, lavage the rats of oxymatrine administration group with oxymatrine during the molding time, while feed physiological saline in same quantity to the rats of the other two groups. Six weeks later, rats were sacrificed and the specimens were routinely processed. Then, periodontal tissues were observed by histology and the distance from cemento- enamel junction to the alveolar bone crest was measured.Results: The periodontal inflammation in the rats treated with oxymatrine was significantly slighter than those were treated with physiological saline, and the bone absorption was relatively slighter (P < 0. 05).Conclusions: This study suggests that oxymatrine can prohibit alveolar bone absorption of periodontitis.2.The effects of oxymatrine on biological behaviour of PDLC under existing LPS.Method: Firstly, detach PDLC externally and cultivate them; for oxymatrine administration group we use different combination of LPS and oxymatrine in different concentration, and for the other two groups we use DEME nutrient fluid of 1percent FBS. Then, MTT method was used to examine PDLC'multiplication situation, Flow cytometry was used to measurate the cycle distribution of PDLC, and transmission electron microscope(TEM) was used to observe the change of PDLC ultrastructure; and ELISA to examint ALP active influence of PDLC; then RT-PCR for checking the level of IL-6 and IL-1βmRNA.Results: LPS can significantly prohibit the proliferation of PDLC, and the percentage of human periodontal ligament fibroblast in G1 phase during exponential growth was increased, while the percentage of human periodontal ligament fibroblast in S phase was decreased. The ALP activity was reduced, the ultrastructure of PDLC was injured, and the expression of both IL-6 and IL-1βmRNA's level were boosted up by LPS, but oxymatrine could ameliorate above phenomenas.Conclusion: Oxymatrine can interrupt LPS's damage to PDLC in some extent, prohibit the delivery of inflammation medium, and provide protection to PDLC.
