PURPOSE: Retina ischemia-reperfusion injury is a frequent clinical pathological process in ophthalmology. It can be taken place in central retinal artery occlusion ( CRAO ) , ischemic optic neuropathy ( ION ), glaucoma and so on. As an important objective index evaluating retinal function, b wave is now increasingly applied in the examination. It represents electric activity of inner nuclear layer of retina. b wave, which is a sharply ascending large and positive wave following a wave, is the PII ingredient of Granit three lead concepts. The amplitude of b wave after dark adaptation is lower than that of light adaptation. b wave can be easily affected by oxygen deficiency, temperature change, prospective adaptable state and so on. Visual evoked potential(VEP) test is of great value in visual pathway diseases. It is not only important in understanding the changes of different visual diseases, offering the information about the extent of degeneration extent and medullary sheath; but also important in the disease diagnosis estimation and prognosis. Patients who have optic neuropathy have the abnormality in fundamental wave and amplitude of vibration swing when faced 30Hz flicker. The total abnormality ratio can arrive at 80.4%.The experiment wants to observe the protection of verapamil in retina and optic nerve by HE dyeing. On the other hand, observe the effect of verapmil on b wave and P wave by visual electrophysiology.METHODS: 1. 48 Sprague-Dawley(SD) rats were randomly divided into two groups: Verapamil administrated group(group I, n=24) and the Isotonic Na chloride group (group II , n=24).Choose the left eye as the retina ischemia-reperfusion injury eye. Rats are received ERG and VEP test after one hour, six hours, twelve hours, forty-eight hours and seventy-two hours. 2. Observe the retinal changes by HE dyeing slice.RESULTS: 1.Successfully build the acute retinal ischemia model by paracentesis of anterior chamber.2.It is obvious of the morphologic changes of the retina after retina ischemia-reperfusion injury. 3.6 hours after reperfusion, retina b wave recover better in verapamil graph than in contrast graph. Verapamil enhance ERG-b and VEP-P wave of retina after ischemia-reperfusion injury.CONCLUSION: 1. Verapmil benefit to the recovery of b wave 2.Verapmil benefit to the recovery of P wave .3.Verapmil can obviously alleviate the damage of retina after ischemia-reperfusion injury...
