ObjectiveTo observe the expression of vascular cell adhesion molecule1(VCAM-1) and basic fibroblast growth factor(bFGF) in renal tissue of hyperlipidemia rabbits and to study the renal protective effect of atorvastatin.MethodsTwenty-four male New Zealand white rabbits were divided into three groups randomly,eight rabbits per group:control group(Group A),high fat diet group(Group B),atorvastatin treated group(Group C).After ten weeks,collecting the empty stomach blood preparation through central artery at awake state(fasting diet twelve hours), centrifugalization(1000r/min,10min), blood serum was stored at -20℃and the levels of total cholesterol(TC),triglyeride(TG) and low-density lipoprotein(LDL) were determined by the BackmaanLX20 automatic biochemistry analyzer;The renal pathology change was observed by HE staining and the levels of the expression of VCAM-1 and bFGF were determined by immunohistochemistry.Results1)The TC,TG and LDL levels of the three groups:The TC level of high fat diet group is increased significantly compared to control group(21.08±1.25 vs 2.21±0.45,P<0.05);The TC level of atorvastatin treated group is decreased significantly compared to high fat diet group(8.16±0.28 vs 21.08±1.25,P<0.05).The TG level of high fat diet group is increased significantly compared to control group(6.83±0.64 vs 1.18±0.48,P<0.05);The TG level of atorvastatin treated group is decreased significantly compared to high fat diet group(2.37±0.33 vs 6.83±0.64,P<0.05). The LDL level of high fat diet group is increased significantly compared to control group(15.84±0.76 vs 1.17±0.27,P<0.05); The LDL level of atorvastatin treated group is decreased significantly compared to high fat diet group(6.27±0.31 vs 15.84±0.76,P<0.05).2)The expression of VCAM-1 and bFGF in glomcrulus of the three groups:The expression of VCAM-1 of high fat diet group is increased significantly compared to control group(9.87±1.20 vs 1.59±0.55,P<0.05);The expression of VCAM-1 of treated group is decreased significantly compared to high fat diet group(4.18±0.91 vs 9.87±1.20,P<0.05). The expression of bFGF of high fat diet group is increased significantly compared to control group(12.21±1.80 vs 0.42±0.23,P<0.05); The expression of bFGF of treated group is decreased significantly compared to high fat diet group(5.38±0.82 vs 12.21±1.80,P<0.05).3) The expression of VCAM-1 and bFGF in nephric tubule and interstitium of the three groups: The expression of VCAM-1 of high fat diet group is increased significantly compared to control group(13.54±1.30 vs 1.50±0.68,P<0.05);The expression of VCAM-1 of treated group is decreased significantly compared to high fat diet group(5.82±1.07 vs 13.54±1.30,P<0.05); The expression of bFGF of high fat diet group is increased significantly compared to control group(11.76±1.62 vs 0.37±0.36,P<0.05); The expression of bFGF of treated group is decreased significantly compared to high fat diet group(3.53±1.11 vs 11.76±1.62,P<0.05). ConclusionThe levels of the expression of VCAM-1 and bFGF were significantly increased in renal tissue of hyperlipidemia rabbits;Atorvastatin could lessen the renal inflammatory reaction,inhibit the renal fibrosis by reducing the expression of VCAM-1 and bFGF in renal tissue, accordingly,retard the chronic renal failure to some extent.
