Objective:To investigate in vitro adhesion and colonization of streptococcus sobrinus incubated with IgY toothpaste in vivo in rats.Methods:Experiment One:①S.sobrinus were cultured with different concentrations of IgY (2.5mg/ml, 1.25 mg/ml,0.625 mg/ml,0.3125 mg/ml), IgY toothpaste and control toothpaste respectively. After culture,the adhesion of S.sobrinus was observed by spectrophotometer.②Cultured streptococcus sobrinus treated with anticarious IgY and IgY toothpaste was observed by transmission electron microscope(TEM).Experiment Two: 24 Wistar rats were randomly divided into 3 groups. All rats were fed with sterilized water containing Ampicilin 0.25㎎/ml at the first week and incubated with S.sobrinus 6715 twice a day at the second week.At the third week,different solution were smeared on the surface of the molars for three weeks. Bacterium samples were scraped in order to determine the numbers of S.sobrinus by MS plates and general bacterium by BHI plates.Then measured the proportion of S.sobrinus to gross colony .Results:Experiment One:①The adhesion rate had significantly difference between IgY toothpaste and control toothpaste(P<0.05),but no significantly difference between IgY toothpaste and egg yolk IgY(0.3125 mg/ml) (P>0.05).②The capsule of cell wall was dissolved in anticarious IgY group. In IgY toothpaste group some cells were dissolved,other than the above changes.Experiment Two:The proportion of S.sobrinus to gross colony had significantly difference between IgY toothpaste and control toothpaste(P<0.05),but no significantly difference between IgY toothpaste and egg yolk IgY(0.3125 mg/ml) (P>0.05).Conclusion: IgY toothpaste can inhibit in vitro adhesion of S.sobrinus. IgY toothpaste could lead to the loss of the cell capsule of streptococcus sobrinus and influences the adhesive effect.IgY toothpaste can inhibit in vivo colonization of S.sobrinus.
