Objective To investigate the expression and role of nuclear factor-κB(NF-κB),tumor necrosis factor-α(TNF-α) and intercellular adhesion molecule-1(ICAM-1) in human idiopathic macular epiretinal membrane and macular epiretinal membrane in diabetic retinopathy and retinal detachment.To discuss the pathogenesis of human macular epiretinal membrane.Methods The experiment was divided into three groups,which were idiopathic macular epiretinal membrane group,macular epiretinal membrane in diabetic retinopathy group and mac -ular epiretinal membrane in retinal detachment group.Inner limiting membranes of normal mac -ular tissue was detected in control group.The expression of NF-κB,TNF-αand ICAM-1 was observed by fluorescein immunohistochemical methods in 12 human idiopathic macular epiretin -al membrane,16 human macular epiretinal membrane in diabetic retinopathy,18 human macul -ar epiretinal membrane in retinal detachment and 9 inner limiting membrane of normal macular tissue.The morphological character of macular epiretinal membrane was observed by HE.Results Expression of NF-κB,TNF-αand ICAM-1 were respectively observed in 8,10 and 9 idiopathic macular epiretinal membrane,positive rate were 66.7%,83.3%and 75%. Expression of NF-κB,TNF-αand ICAM-1 were respectively observed in 12,13 and 10 macular epiretinal membrane in diabetic retinopathy,positive rate were 75%,81.3%and 62.5%. Expression of NF-κB,TNF-αand ICAM-1 were respectively observed in 11,15and 13 macular epiretinal membrane in retinal detachment,positive rate were 61.1%,83.3%and 72.2%.No positive staining was found in the inner limiting membrane of normal macular tissue.The data was analyzed by partitions of x~2 and ANOVA.All experiment groups were significantly different compared to control group in the same condition(p<0.01).The expression of NF-κB with TNF-αand ICAM-1 were analyzed by pearson linear correlation.The expression of NF-κB and TNF-αhad significantly positive correlation in idiopathic macular epiretinal membrane(r=0.961,p<0.01),but had no correlation with ICAM-1 in idiopathic macular epiretinal membrane(r=-0.240,P=0.453).The expression of NF-κB with TNF-αand ICAM-1 had significantly positive correlation in macular epiretinal membrane in diabetic retinopathy (r=0.614,P=0.011.r=0.686,P=0.003).The expression of NF-κB with TNF-αand ICAM-1 had significantly positive correlation in macular epiretinal membrane in retinal detachment (r=0.833,P<0.01.r=0.905,P<0.01).Conclusion(1) The findings indicated that NF-κB,TNF-αand ICAM-1 mediated pathways of inflammation are involved in the pathogenesis of human macular epiretinal membrane in human idiopathic macular epiretinal membrane and macular epiretinal membrane in diabetic retinopathy and retinal detachment.(2) It was confirmed that NF-κB and TNF-αmediated pathways existed in idiopathic macular epiretinal membrane.The expression of ICAM-1 may exist other signal pathways,while ICAM-1 itself was related with inflammation.(3) It was existed NF-κB,TNF-αand ICAM-1 mediated pathways of inflammation in the pathogenesis of macular epiretinal membrane in diabetic retinopathy and NF-κB,TNF-αand ICAM-1 had cooperated function and had positive correlation.(4) It was existed NF-κB,TNF-αand ICAM-1 mediated pathways of inflammation in the pathogenesis of macular epiretinal membrane in retinal detachment.NF-κB,TNF-αand ICAM-1 had cooperated function and had positive correlation.
