Cytostasis Action Study Of Sorafinib On Pancreatic Carcinoma Cell

Background/purpose:Investigating the effect of catastasis and induction apoptosis action on sorafinib to pancreatic carcinoma cell line BXPC-3。Method:Inverted microscope observe the morphologic change of cell;Pancreatic carcinoma cell line BXPC-3 were cultured in vitro.MTT assay were used to determineted the antiproliferative effect of different doses sorafinib on pancreatic carcinoma cell BXPC-3 and inhibition ratio were calculated. Cell cycle in the pancreatic carcinoma cell BXPC-3 after dfferent doses sorafinib were evaluated by flow cytometry.Result:Inverted microscope observe contiguously, along with temporal prolongation,cell appear shrinkage,round,abscission.The pancreatic carcinoma cell BXPC-3 after different doses sorafinib, along with temporal prolongation, growth curve of cell became low and equal, even appear reverse direction. along with concentration increasing, growth curve of cell became low and equal. After diffent doses sorafinib (10,5,2.5,1,0.5μmol/L)and pancreatic carcinoma cell line BXPC-3 co-cultivation,MTT assay detection, after 48 hours ,cell surviva and growth circumstance detection.the OD value,cell survival rate,cell inhibition rate comparison of each concentration group and control group have statistical significance(P<0.01). Along with concentration increasing, cell inhibition rate to increase gradually,for: 26.1%,40.52%,56.06%,70.4%,78.44%. to compare with control group: 10μmol/L group,5μmol/L group and 2.5μmol/L group have significance(P<0.01). otherwise groups without significance(P>0.05). After different doses sorafinib effect, flow cytometry analyze the different circumstance of cell cycle distribution. After sorafinib effect 24 hours,along with concentration increase, G0/G1 stage cells proportion are: 64.56%,58.79%,54.55%,47.88%,45.31%. drug concentration over 2.5μmol/L, G0/G1 stage show raise up markedly,having statistical significance(P<0.05),also G2/M stage have slight increase, discripting sorafinib block BXPC-3 cell in G0/G1 stage,also G2/M stage have a epactal block. Meanwhile before G0/G1 stage,it appear typical hypodiploid apoptotic peak(AP peak).Conclusion:1.sorafinib can inhibit hyperplasy of pancreatic carcinoma cell line BXPC-3. 2. sorafinib have action of inducing apoptosis. 3. sorafinib can block pancreatic carcinoma cell line BXPC-3 in G0/G1 stage.
