Objective: To observe the expression and its change regularity of nuclear factor-kappa B(NF-кB) following experimental traumatic brain injury (TBI) in rats, and investigate the protective effect of pyrrolidine dithiocarbamate (PDTC).Methods:The model of local moderate brain contusion was established by the free-falling-body impact device.96 rats were randomly devided into three groups :TBI group(T group)(n =72),TBI+PDTC prevetion group(P group)(n=72),and sham operation group (S group) (n =24).Then T group was randomly devided into three groups: mild TBI group (T1),midrange TBI group (T2),severe TBI group (T2), and P group was also devided into three groups:mild TBI+PDTC group (P1),midrange TBI+PDTC group (P2),severe TBI+PDTC group (P3).Each group was devided into 2 hour ,12 hour ,24 hour ,48 hour ,72 hour subgroups . PDTC was ip given after TBI in P1,P2 and P3(100mg/kg),and the other groups was ip given chloride . The expression of NF-кB were detected by using RT-PCR at 2h,12 h,24h,48h respectively after TBI.Results:1,The expression of NF-кB in the area surrounding the injured brain were low in the sham-operated group, while which increased from 2h,continued to raise up at 12h ,reached the summit at 24h, and decrease at 48h . Significant difference was found among the S group and T group(P<0.05); 2,PDTC inhibited the expression of NF-кB ,and significant difference was also found among the P group and Tgroup(P<0.05).There was no significant differrence among the S group and P group(P>0.05); 3,The expression of NF-кB was different at the same time point(except for 2h) among T1,T2 and T3 groups,and the difference was most significant at 24h : The expression of NF-кB increased with injury degree. There was no significant differrence at the same time point among P1,P2 and P3 groups(P>0.05). 4,Significant difference was found between the former time point and the latter in T1,T2 and T3 groups(P<0.05),and which was opposite in P1,P2 and P3 groups(P>0.05).Conclusion : 1,The expression of NF-кB in the area surrounding the injured brain in rats after TBI increased,.and reached the summit at 24h. The expression of NF-кB was not only intimate with the time after TBI,but also it was relevant with injury degree .At the expression summit the expression of NF-кB increased with injury degree..The activation of NF-кB contributes to the secondary brain injury. 2,Using PDTC after TBI can effectively inhibit the expression of NF-κB and therefore protect the brain tissue after TBI.
