Background: Research has been confirmed at domestricity and abroad, apoptosis to participate in tumor is in a very close relationship, apoptosis and uncontrolled proliferation of cells is one of the important factors of tumor development. The occurrence of tumor has close relationships not only with the over expression of oncogene that control cell proliferation, but also with inactivation and mutation of oncogene that inhibit apoptosis and induct apoptosis in cancer cell. The mechanism of apoptosis that was inappropriate termination is an important prerequisite to cancer cells, and the inhibition of apoptosis in tumor development plays an important role. In 1972, Kerr put forward the concept of apoptosis chiefly. The concept that was also called programmed cell death is a mechanism that was involved a lot in the process of cell organisms removal, and it is initiative process of cell suicide that programmed by genes. Through the mechanism, the organisms remove the aging and abnormal cells. In addition, apoptosis has a important impact on embryo development, immune tolerance, cell groups'stability, and apoptosis has a potential value for the further study of AIDS, cancer that pose a serious threat to the survival of mankind. Therefore, cell apoptosis has been a hot spot in the area of scientific research on biology for many years. Apoptosis process is very complicated, in connection with which two families to consist of Bcl-2 and CasPase-3 play an important role on the regulation of apoptosis. CasPase-3 is a group of structure related cysteine protease that existed in the cytoplasm Sol, in which an important common ground specific is disconnect between aspartate residue peptide bond. The word"CasPase"was derived from the abbreviation of the first initials of the phreath Cysteine aspartic acic specific protease. So far, 14 kinds of CasPase enzymes of mammals have been found, witch were in prodormant state in common condition, but under some specific conditions, these enzymes are processed into the activity mature enzymes. Based on the substrate specificity and cell function, CasPase can be divided into three groups:â… the inflammation group: CasPase1, 4,5,11,12,13,14.â…¡the group of apoptosis to Start: CasPase6, 8, 9 and 10.â…¢the group of apoptosis effectiveness: Caspase2, 3, 7. The occurrence of apoptosis is a complicated protease cascade process that is guided by the CasPase family groupe, although which apoptosis is induced by different cells or different signal transduction pathway are in some different on participate CasPase, but CasPase-3 is the key enzyme in apoptosis cascade reaction. Bcl-2 family in the process of apoptosis play the role regulator, the family is divided into three sub-group: the Pro-survival subfamily that the Bcl-2 family members of Bcl-2 and BCL-CL, KS- Bcl-2 and BCL-W, MCL-1, BHRF1, NR - B, ORF16, LMW5-HL, AL, FIB-19K, and CED-9; Two original family of apoptosis (Proapoptotrc subfamily) are Bax and BH3 subtribe. Bcl-2 gene (B-cell lymphoma / leukemia-2 gene) is a proto-oncogene, it could inhibit apoptosis. The role sites of Bcl-2 is exist between the signal molecule and effect position in the apoptosis pathway.A lot of research have show that Bcl-2 and CasPase-3 inhibition each orther, Bcl-2 inhibited the intensification of CasPase-3.The active CasPase-3 can biodegradate protein Bcl-2, to make its expression decline. At present, the Chinese medicine that induced apoptosis in tumor cells mainly concentrated in the molecular regulation mechanisms. YiMiHuaJiYiGanJian is the traditional Chinese medicine for anti-cancer, especially for primary liver cancer and metastatic cancer. In view of this, the experiment through molecular biology methods to detect the expression of CasPase-3 and Bcl-2 to study the mechanism that inhibit the tumor of armpit entities tumor in mouse model and the anti-tumor molecular mechanism of Chinese herbal compound. Objective: Through the application of Chinese medicine YiMiHuaJiYiGanJian inhibit liver cancer growth and metastasis in tumor-bearing mice to detect the level of CasPase-3 and Bcl-2l to study the anti-tumor molecular mechanism of Chinese herbal compound.Method: With randomized method categorized mice into 4 groups, including normal control , bearing cancer control group, Chinese medicine treatment group and 5-fluorouracil chemotherapy treatment group, then establish solid tumors in mice armpit tumor model. Under test immuno-histochemical assay was used to detect the expression of CasPase-3 and Bcl-2l to study the anti-tumor molecular mechanism of Chinese herbal compound. Last analyze the significance of data.Result:The expression of CasPase-3 in Chinese medicine treatment group and 5-fluorouracil chemotherapy treatment group were increased, and the significant difference was found between the two groups and normal control group(P<0.01); the expression of Bcl-2 in Chinese medicine treatment group and 5-fluorouracil chemotherapy treatment group was higher than that of other groups; The significant difference of the expression of CasPase-3 and Bcl-2 in Chinese medicine treatment group and 5-fluorouracil chemotherapy treatment group was not found(P<0.05)Conclusion: The anti-tumor effect of Chinese medicine YiMiHuaJiYiGanJian is through the mechanism of apoptosis, raised CasPase-3 protein expression and lower levels of Bcl-2 protein expression levels to promote apoptosis of tumor cells, so that a cancer tumor necrosis role.
