Objective: To investigate the effect of prostaglandin E1(PGE1)on inflammatory cytokines expression in kupffer cell (KC) induced by lipopolysaccharides (LPS) and also to observe the change of the TNF-αand nuclear factor-kB (NF-kB) secreted by KC, which are stimulated by LPS and PGE1.To investigate the anti-inflammatory action and mechanism of PGE1 and the result may give us some new ideas for clinic use.Methods: KCs were isolated from livers of healthy male adults SD rate and was cultured for 12h. The cultured cell, concentrating 5x105, are inoculated into the 24 holes culture medium.The isolated KCs were cultured for 48h. Then, toxicity was evaluated by determination of LDH activity 3 hours after PGE1 (10-100μmol) administration.KCs were normally cultured or incubated with LPS or LPS plus PGE1 (3 different concentration within safe scale), and were stimulated in 3h. Investigate the change of the releasing of TNF-αand IL-6 and the activity of the NF- kB.The expression of TNF-αand IL-6 in the culture supernatant was assayed by enzyme-linked immunosorbentassay (ELISA) and radioimmunoassay (RIA). We determinate the NF-kB activity in KCs by electrophoretic mobility gel-shift assay (EMSA)Results: The level of NF-kB, TNF-αand IL-6, amylase in serum of stimulating group were significantly higher than those of contrasting group (p<0.01). The expression of TNF-αand IL-6 in KCs remarkably decreased after PGE1 administration in dose-dependent manner (p<0.05).Conclusion: When the liver were injury by various acute or chronic diseases. LPS enable amounts of TNF-αand IL-6 through the activation of NF-kB. During the disease, many factors have very important effects, such as NF-kB activity which start up the voluming expression of TNF-αand IL-6, excessive activation of KCs, a large amount of inflammatory factors, higher level of LPS in blood, et al. PGE1 could exert protecting effects through eliminating NF-kB activity, reducing excessive activation of KC, alleviating the mass expression of TNF-αand IL-6,decreasing the higher level of LPS, et al.
