Objective:This study was to show the effects of family allergic history and allergen stimulation over the excursion of healthy neonate Th1/Th2 cell,by testing the expression of CBMC Th1/Th2 cytokines.Methods:We got 30 samples of healthy newborn baby cord blood(each sample are 4ml),half of which are from the babies with family allergic history as testing group; others with no family allergic history as control.CBMCs were isolated by Ficoll gradient centrifugation.Then we divided into no-allergen stimulated group,PHA stimulated group and different concentration of Dermatophagoides farinae(Der f) stimulated group and cultured these CBMCs for 48h.The concentrations of IFN-γand IL-4 in the culture supernatant were determined with ELISA kits according to the manufacturer's instructions.Results:(1)In no-allergen stimulated group,the concentrations of IL-4 in the culture supematant was 11.35±1.80 ng/L in the testing group and 11.00±1.50 ng/L in the control group,t=0.576,p>0.05,there was no significant difference.The concentration of IFN-γwas 9.55±1.47 ng/L in the testing group and 10.19±1.39 ng/L in the control group,t=-1.235,p>0.05,and there was no significant difference.(2)In PHA stimulated group,the concentrations of IL-4 in the culture supematant was 43.45±4.57 ng/L in the testing group and 37.58±3.41 ng/L in the control group,t=3.992,P<0.001, and there was significant difference.The concentration of IFN-γwas 72.61±25.40 ng/L in the testing group and 65.84±29.96 ng/L in the control group,t=0.667,p>0.05,and no significant difference.(3)In low density Dermatophagoides farinae stimulated group, the concentrations of IL-4 in the culture supematant was 40.54±3.64 ng/L in the testing group and 37.17±2.60 ng/L in the control group,t=2.881,P<0.01.There was significant difference.The concentration of IFN-γwas 35.30±2.73 ng/L in the testing group and 40.55±1.85 ng/L in the control group,t=-6.108,P<0.001.There was significant difference.(4)In high density Dermatophagoides farinae stimulated group,the concentrations of IL-4 in the culture supematant was 43.50±3.19 ng/L in the testing group and 39.55±4.13 ng/L in the control group,t=-2.860,P<0.01.There was significant difference.Thr concentration of IFN-γwas 39.40±5.21 ng/L in the testing group and 40.94±2.96 ng/L in the control group,t=-0.989,P>0.05,and no significant difference. We found that allergic diseases were happened in 7 cases of 13 cases with family allergic history and in 2 cases of 15 cases without family allergic history in one year fellowing up of two groups.There was significant difference between the two groups.Conclusions:Family allergic history has effect on the balance of neonate Th1/Th2 cells.In the group of healthy newborn baby with family allergic history,the secretion of IL-4 expression level of Th2 increased significantly,which means Th2 cells were dominant.This phenomenon implies that there may be a relationship between the Th2 domination in neonate period and the fact that the occurrence of allergic diseases were easily in children with family allergic history.
