ObjectiveTo calculate the ratios of CD4~+CD25~+T cells and CCR4~+cells to lymphocytes in Tumor Infiltrated Lymphocytes(TILs)and peripheral blood of nasopharyngeal carcinoma,then to probe the immune function and whether CCR4 play a role in tumor immunity,so as to provide theorial basis for clinical therapy.MethodsThe ratios of CD4~+CD25~+T cells and CCR4~+cells to lymphocytes were measured by FCM in TILs(25 cases)and peripheral blood(35cases)from nasopharyngeal carcinoma patients,None of those had undergone radiotherapy or chemotherapy.The same method did for tissue and peripheral blood from control group(normal in12cases).Then we did the statistical analysis, stimulated the relationship between CD4~+CD25~+T cells and CCR4 and the immune function of different stages according to clinical information,explored how the CCR4 play a role in help CD4~+CD25~+T cells to enhance the immunosuppression.ResultsThe ratios of CD4~+CD25~+T cells in TILs and peripheral blood of NPC were higher than control group(just as 16.19±7.78%vs 3.57±1.20%,5.43±3.32%vs 3.24±1.34%,p<0.05 );consistently CCR4~+cells level were elevated in TILs and peripheral blood of NPC comparing to in control group(47.09±29.77%vs 2.62±1.12%,11.22±7.41%vs 2.43±1.89%,p<0.05);the ratios of CD4~+CD25~+T cells and CCR4~+cells in tissue were higher than in peripheral blood respectively(p<0.05),but there no so difference between tissue and peripheral blood in control group(p>0.05);The ratio of CD4~+CD25~+T cells in TILs at stageⅢ+Ⅳwas higher than the stageⅠ+Ⅱof NPC(23.76±12.29%vs 2.70±5.76%,p<0.05 ),but there was no so difference between the two stage in peripheral blood in NPC;there was positive correlation between CD4~+CD25~+T cells and CCR4~+ cells in TILs and peripheral blood of NPC(correlation coefficient is 0.85,0.78,p<0.05);and There was negative correlation between CD4~+CD25~+Tcells and CD4~+CD25~- T cells in TILs of NPC(correlation coefficient is-0.75,p<0.05).ConclusionsThe ratio of CD4~+CD25~+T cells was higher both in TILs and peripheral blood in NPC,which showed NPC patients are in immunosuppression,even so significantly elevated in TILs as to regard there is more severe immunosuppression in tumor sites;the ratio of CD4~+CD25~|T cells was correlated with the stages of NPC,so the NPC progress to later stages,the percentage of CD4~+CD25~+T cells increased which was correlated with poor prognosis;the ratio of CD4~+CD25~+T cells in NPC was positive correlated with the percentage of CCR4~+ cells,which suggested CCR4 played an important role in reactant of CD4~+CD25~+T cells to tumor sites,and resistant to immunosurveillance.
