Objective To investigate the relation of genetic polymorphisms ofβ2 adrenoceptor (β2AR)at codons 16 and 17(A rg16Gly+Gln27Glu)on longacting bronchodilator response in patients with COPDMethods All patients with COPD that initially numbered 126 individuals who were randomized underwent assessments of lung function;genotyping for polymorphisms of the beta(2)AR at codons 16 and 17 was performed. After one month, these patients who have inhaled formoterol were tested lung function again. To compare relation of differentβ2AR genetic polymorphism and formoterol bronchodilator response. Results Patients who had homozygous or heterozygous genotypes containing the arginine16 polymorphism(Arg16/Arg16(n=37),Arg16/Gly16(n=41))had lower bronchodilator response after inhaling formoterol(n=19 and n=21 respectively), however, the homozygous glycine16 genotype Gly16/Gly16(n=48)had a significant greater bronchodilator response(n=41 P<0.05).But no independent associations were observed between theβ2AR genetic polymorphism in 27 position and bronchodilator response.Meanwhile, the most common Arg16Gln27 haplotypes reduced bronchodilator response remarkably(P<0.01).Conclusionsβ2 adrenoceptor genetic polymorphisms may influence longactingβ2 adrenoceptor agonist reactivity. So the clinical effects of different longactingβ2 adrenoceptor agonist can be explained.
