Objective: To study the distributing characteristics of D2-40 or CD34 positive vessels in colorectal carcinomas (CRC) and discuss the correlated significance of microlymphatic and microvascular density and proliferative activity of the endothclium of lymphatic vessels and blood vessels with lymph mode metastasis. To study the expression of VEGFR-3 in the lymphatic vessel and blood vessel endotheliocyte and to investigate the relationship of the expression between VEGFR-3 and VEGF-C and discuss the significance of them with lymph node metastasis in human colon carcinomas.Methods: With 50 specimens of human colonic carcinoma and normal tumor neighboring mucosa, by SP immunohistochemistry with monoclonal antibodyes against CD34,D2-40 and KI-67, to investigate the distribution patterns of microlymphatic vessel and microvacu -lar vessel and proliferative activity of the endothclium of lymphatic vessels and blood vessels in colorectal carcinomas (CRC) and to study their relationship with tumor metastasis. By SP immunohistochemistry with antibodyes against VEGFR-3 and CD34,to study and research expression of VEGFR-3 in microlymphatic vessel and microvacular vessel; By SP immunohistochemistry with polyclonal antibodyes against VEGF-C,VEGFR-3,to study their expression in 50 specimens of human colonic carcinoma and normal tumor neighboring mucosa and investigate their relationship with tumor metastasis. During SP immunohistochemistry staining set up positive control and negative controlResults: D2-40 and CD34 maker of tumor microlymphatic vessel and microvacular vessel, respectively, microlymphatic density (MLD) and microvascular density (MVD)in the different area of tumors could be measured to detect the distribution of microlymphatic vessel and microvacular vessel and find out the relationship of migra -tion and CRC prognosis. We find that D2-40 and CD34 can either be detected in norm -al tumor neighboring mucosa or colorectal cancer, but in total, D2-40(MLD) expression was much lower than CD34(MVD). Similarities exist in the distribution pattern ofD2-40 and CD34 in colorectal carcinoma, that is, both of them are most distributed in tumor in -vasive front, less in superficial zone, least in center part, D2-40 especially, which could -n't be detected at all. It is infered that MLD distribution of tumor invasive front is more intense compared with central region, superficial zone as well as cancer-neighboring mucosa MLD (P<0.01 or P<0.05)by correlation analyses, however, there are no statistic difference between total colorectal cancer MLD and cancer-neighboring mucosa MLD (P>0.05).In addition, statistics refered to tumor's anterior field MVD distribution shows the similar pattern to MLD, which distributed much more than in central region, superfi -cial region and normal cancer neighboring mucosa (P<0.01 or P<0.05).There are no difference between total colorectal cancer MVD and normal cancer neighboringmucosa MVD either (P>0.05).It also can inferred from those tatistics that tumor invasive front MLD was mainly correlative with tumor invasive depth, TNM Stage, vascular invasion, lymphatic node metastasis and distant organ metastasis (P<0.05). MLD/MVD of anterior field highly correlative with metastasis and which were credible indicator of lymph node metastasis, depth of invasion and adverse prognosis for CRC.VEGFR-3 was mainly expressed in the endothclium of lymphatic vessels in the tissue of colorectal carcinomas, but seldom see in the endothelium of the blood vessels shows great diversity between them (P<0.01); VEGFR-3 expression rates in the perimeter tissue of colorectal carcinomas were significently higher than those in the interior (P<0.01). VEGFR-3 positive expression rates were correlated with the density of lymphatic vessel. The VEGFR-3 positive vessel was correlated with depth of invasion, lymph node metastasis and TNM classification (P<0.01 or P<0.05). However, it was not related to sexuality, age, size of tumor, depth of invasion, histological type and distant metastasis (P>0.05) Conclusion:1. The colonrectum microlymphatic and microvascular density is most intensive in the perimeter of tumor, which is a significance indicator in the aspect of depth of invasion and lymph node metastasis; especially, the microlymphatic density and proliferative activity microlymphatic endotheliocyte of perimeter of tumor.2. The co-expression of VEGFR-3 and VEGF-C may play an important role in form of lymphatic vessel .VEGFR-3 positive vessels was correlated with lymph node metastasis and TNM classification.
