Objective:To study the correlation of Val34Leu and Pro564Leu polymorphism in the A subunit of coagulation factorⅩⅢ(FⅩⅢA)with cerebral hemorrhage.Methods:The gene include mutations of Val34Leu and Pro564Leu,which loci in exon 2 and 12 of FⅩⅢA,were amplified by polymerase chain reaction(PCR)in 80 patients with cerebral hemorrhage and 80 peoples with no cerebral hemorrhage as control group.Used the method of restriction fragment length polymorphis(RFLP),The products were digested by restriction enzyme. The genotype was assayed with 3%agarose gel electrophoresis.The questionnaire method used to collect two group peoples' Information including the history of hypertension,diabetes and hyperlipidemia.To Analysis and identify the influence of FⅩⅢA Pro564Leu gene polymorphisms on hypertension,diabetes,hyperlipidemia,two groups had been divided into several groups according to peoples whether with those factors.Results:(1).One.case of Leu34 homozygote and one case of Val34Leu heterozygote were found in the cerebral hemorrhage group,but none of which in the control group.(2).We found 8 cases of Leu564 homozygote and 21 cases of Pro564 Leu homozygote in the cerebral hemorrhage group,and one cases of Leu564 homozygote with 6 cases of Leu564 homozygote in the control group. The allele frequency of Leu564 is higher in cerebral hemorrhage group than in control group.(P<0.05).(3).there were no significant difference that FⅩⅢA Pro564Leu genotype and allele frequency distribut in peoples,whether whom with hypertension,diabetes,hyperlipidemia.(P>0.05)Conclusions:Our results showed that FⅩⅢA Val34Leu gene polymorphis have no correlation with cerebral hemorrhage,but peoples with FⅩⅢA Pro564Leu polymorphisms can increase the fatalness of cerebral hemorrhage. We Found that FⅩⅢA Pro564Leu gene polymorphism have no correlation with hypertension,diabetes or hyperlipidemia.
