| Objective: The study was to determine if PLGA/HA composite could serve as an effective scaffold for cartilage repair and the ability of MSC and PLGA/HA composite in the cartilage repair.Method: MSCs were isolated from the SD rat bone marrow and cultured on the PLGA/HA composite. After seven days, the PLGA/HA composite with MSCs were transplanted into the cartilage defects created in the both distal femurs of SD rats. The rats were taken into three groups with 8 rats each group : control group,PLGA/HA group and PLGA/HA-MSC group. The rats were sacrificed after 6 and 12 weeks and the species were obtained for macroscopic examination and histological evaluation. Immunohistological staining of collagen II was carried out.Result: The control group was repaired with fibrous tissue. The PLGA/HA and PLGA/HA-MSC group were repaired with hyaline cartilage. The score was significantly different between the control group and another two groups. The total score was also significantly different between the PLGA/HA and PLGA/HA-MSC group. Abundance of cartilaginous extracellular matrix and Col II accumulation were observed after implantation of the PLGA/HA composite and PLGA/HA incorporated with MSCs.Conclusion: The PLGA/HA composite was able to repair the cartilage defect and MSCs could enhance cartilage repair. |
