| OBJECTIVE: To observe the change of the expression of FN,CD44 and CD54 in cancellous bone of ovariectomy rat and investigate the mechanism of Exercise and Epimedium Herb in preventing and curing postmenopausal osteoporosis.MATERIALS AND METHODS:34 female wistar rats were divided into four groups at random. Removed both ovaries of all rats.10,as control group,marked as"OVX". 8, as swim group, marked as"S",were do swim exercise after one week after ovariectomy. 8,as medicine group, marked as"M",were given Epimedium Herb cure after one week after ovariectomy.8, as swim and medicine group, marked as"SM", were do swim exercise and given Epimedium Herb cure at the same time after one week after ovariectomy. Two rats of control group were killed at random erery two weeks from the fourth week after ovariectomy.Take both side of knee joint.and the S,M,SM group were killed after twelve weeks after ovariectomy.Take both side of knee joint.Materials were made into paraffin section . Use immunohistochemistry test the expression of FN,CD44 and CD54.Use Image-Pro Plus 6.0 and SPSS15.0 analyse the photos and datas of IOD.RESULTS:1.Lots of polynucleolar macrophage\ osteoclast like cells ,which shew high expression of FN, could be found in bone tissue of OVX group after ovariectomy . compared with OVX-12, the expression of FN in group S,M,SM were lower but had no significant difference (P>0.05).2.After ovariectomy, the expression of CD44 could be found in each sample of group OVX and rose gradually with time went on. The expression of CD44 in group S,M,SM were lower than that in control group (OVX-12) and had a significant difference(P<0.05) .3. Most CD54 expressed on the surface of membrane or in the matrix of polynucleation osteoclast like cells in control group (OVX-12),S,M. Compared with control group , Group S and M had lower expression of CD54 . In group SM, most CD54 expressed on the surface or in matrix of giant cells whose nucleus were not saw clearly. CD54 expressed negative or weakly positive in polynucleation osteoclast like cells.CONCLUSIONS: 1. Exercise ,for one side, may cause down regulation of the expression of CD44 and CD54 on osteoclast by shearing stress and streamingpotential ,which affected the differentiation and fusion of polynucleolar osteoclast and decreased the lose of bone .For another, Exercise may up regulate the concentration of hormone which have effect on bone metabolism ,then caused down regulation of the expression of CD44 and CD54 on osteoclast and affected the differentiation and fusion of polynucleolar osteoclast.2. By the effective element, chromocor , Epimedium combined with the estrpgen receptor(ER) on the osteoblast and osteoclast caused down regulation of the expression of CD44 and CD54 on osteoclast, which can affect the differentiation and fusion of polynucleolar osteoclast and decrease the lose of bone.3. Exercise combined with Epimedium may inhibit the signal transduction of CD54 between osteoblast and osteoclast ,which reduced the stimulant signal of the differentiation of osteoclast and reduced the quantity of osteoclast.That is the reason why Exercise combined with Epimedium can prevent and cure Osteoporosis . |
