【Objective】To investigate the effect of substance P (SP) and its antagonist(an inhibitor of MAPK and a blocker of calcium channel) on proliferation, adhension and migration of gastric cancer cells and explore its mechanisms.【Methods】The fourth generation of cultured cells were used in the study, gastric cancer cells were cultured in RPMI1640. 100nmol/L SP were added into the RPMI1640,after incubation with SP, we investigate the effect of SP on proliferation, adhension and migration of gastric cancer cells and the changes of MMP-9 secretion of gastric cancer cells. Additionally, we investigate whether the proliferative response of gastric cancer cells to SP were modulated by preincubation with PD-98059(an inhibitor of MAPK ) and Nicardipine(a blocker of calcium channel).【Results】100nmol/L SP can significantly induce proliferative, adhensive and migrative response of the gastric cancer cells, and can promote MMP-9 secretion of the gastric cancer cells, the effect of SP was significantly blocked by PD-98059 and Nicardipine.【Conclusions】 1,SP could effectively promote proliferation, adhesion and migration of gastric cancer cells.2,MAPK signal Inhibitor and calcium channel blocker could inhibit proliferation, adhesion and migration of gastric cancer cells.3,MAPK signal pathaway and culcium signal pathaway played an important role in proliferation,adhesion and migration induced by SP.4,MMP-9 secretion of gastric cancer cell played an important role in migration induced by SP.5,MAPK signal Inhibitor and calcium channel blocker have potential anticancer activity.
