Objective:To investigate the correlation between PTEN,P27 and cyclin D1 expression in colorectal carcinoma and compared it with that of tumor-sideward mucosa and that of normal colorectal tissues,to find the relationship between expression of PTEN, P27 and cyclin D1 in colorectal carcinoma and the pathological features of colorectal carcinoma.Method:Immunohistochemistry was applied to examine the expression of PTEN,P27 and cyclin D1 in 58 cases of human colorectal carcinoma,58 cases of tumor-sideward mucosa and 14 cases of normal colorectal tissues. Results were compared with clinical pathologic parameter such as histology type, lymphatic metastasis and clinical stage.Result:1.Positive expressive rates of PTEN and P27 protein in colorectal carcinoma were 65.52%and 53.45%,which were lower than those in tumor-side ward mucosa and normal colorectal mucosa.Positive expressive rate of cyclin D1 in colorectal carcinoma was 60.34%,which was higher than that in tumor-side ward mucosa and normal colorectal mucosa. The expression of PTEN,P27 and cyclin D1 in colorectal carcinoma were associated with degree of differentiation of carcinoma,Dukes stage and lymph node metastasis of the tumors.2.In colorectal cancer positive expressive rates of PTEN,P27 and cyclin D1 were 65.52%,53.45%and 60.34%respectively,without statistic difference(P>0.05),and correlated with each other(P<0.025),Pearson contingency of coefficient(r_p) =0.8744,0.7135,0.9144.Conclution:The abnormal expression of PTEN,P27 and cyclin D1 participate the malignant conversion process of the colorectal mucosa cell and correlates with tumor stage, histological poor differentiation and lymph node metastasis;PTEN,P27 and cyclin D1 gene play synergistic action in the occurrence and development of colorectal carcinoma, the expression levels of the PTEN,P27 and cyclin D1 protein can be used to evaluate the pathologic biology behavior of colorectal carcinoma.
