ObjectivesTo investigate the efficacy of vitamineE on climacteric ovarian function at various dosage levels for the rat.We aim to obtain understanding on the regulating effect and mechanism of vitamineE on climacteric ovary function.Methods34 12-18 months old female wistar rats were randomized into the following four groups. Group A:(n=8)climacteric rats was treated with vitamine E in high dosage 60mg/kg.d for 4 weeks;Group B:(n=9)climacteric rats was treated with vitamine E in middle dosage 30mg/kg.d for 4 weeks.Group C:(n=9)climacteric rats was treated with vitamine E in low dosage 15mg/kg.d for weeks.Group D:(n=8)climacteric group,was treated with distilled corn oil for 4weeks.Group E:(n=6)Young group,4 months-old,was treated with distilled corn oil for 4weeks.4weeks later,the rats were killed and uteruses and ovaries were weighted separately.Blood samples were collected for measure serum FSH,LH,P and E2.The uterus and ovary were fixed into formalin for histopathology analysis.Young rats were also measured at the same time and compared with the model and treatment groups.Results1.FSH:①The level of FSH in blood serum of the control group(group D:3.81±0.77U/l)is higher than that in the young group(group E2.05±0.59U/L)(P<0.01).②high-dose group(group A2.46±0.58U/L),middle-dose group(group B3.08 ±0.81U/L)and low-dose group(group C2.83±0.79 u/L)are all lower than that in group D.(P<0.05).③The FSH level of group A is a little lower than group B and group C,but there is no significant difference,middle-dose group (group B3.08±0.81U/L)is a little higher than low-dose group(group C2.83±0.79 U/L,),but there is no significant difference.④The FSH level of group A is a little higher than group E,but there is no significant difference.The FSH level of group group B and group C are all higher than group E(P<0.05).2.LH:①The level of LH in blood serum of group D:(7.06±2.29mIU/ml)is a little higher than that in group E(5.50±0.76 mIU/ml),but there is no significant difference..②The LH level of group A(4.26±1.78u/L)is lower than groupD (7.06±2.29 mIU/ml)(P<0.05).group B(6.91±2.84 mIU/ml)and group C5.45±2.12 mIU/ml)are a little lower than groupD(7.06±2.29 mIU/ml),but there is no significant difference.③The LH level of group A is a little lower than group B and groupC,but there is no significant difference. group B is a little higher than group C,but there is no significant difference.④The LH level of group A,groupB and group C are all higher than group E,but there is no significant difference.3.E2:①The level of E2 in blood serum of group D(18.23±6.17pg/ml)is lower than that in group E(34.16±12.53 pg/ml)(P<0.01)。②The E2 level of group A(29.43±9.79 pg/ml)is higher than group D(18.23±6.17pg/ml)(P<0.05), The E2 level of group B(25.4±11.78 pg/ml)and group C(21.06±8.72 pg/ml) are a little higher than group D(18.23±6.17 pg/ml),but there is no significant difference.③The E2 level of group A is a little higher than group B and group C,but there is no significant difference..④The E2 level of group C is lower than group E(P<0.05).4.P:①The level of P in blood serum of group D(7.06±2.29 ng/ml)is lower than that in group E(23.52±9.25 ng/ml)(P<0.01).②The P level of group A(13.05±5.36 ng/ml)is higher than group D(P<0.05),group B(11.24±5.30 ng/ml)and group C(11.79±5.66 ng/ml)are all a little higher than group D,but there is no significant difference.③The P level of group A is a little higher than group B and group C,but there is no significant difference.④The P level of group A,B,C is lower than group E(P<0.01).5.Ovary index:①The ovary index of group D(0.036±0.009)is lower than that in group E(0.059±0.006)(P<0.01).②The ovary index of group A(0.041±0.005),group B(0.039±0.004μ)and group C(0.038±0.005)are all a little higher than group D(0.036±0.009)but there is no significant difference.③There is no significant difference among the ovary index of group A,B,C.④The ovary index of groupA,B,C are all lower than that in group E(P<0.01).6.uterine index.:①The uterus index of group D(0.205±0.047)is lower than that in group E(0.059±0.006)(P<0.05).②The uterus index of group A(0.235±0.113),group B(0.231±0.061)and group C(0.206±0.072)are all a little higher than group D(0.205±0.047)but there is no significant difference.③The uterus index of group C is lower than that in group E(P<0.05).Conclusion:Vitamine E can prevent climaterium rat from retrograde changes of ovary.Efficacy of Vitamine E treatment is concerned of dosage.Vitamine E can improve the numbers of the follicles in the climacteric rats.
