Part 1 Experimental study on thin-layer chromatography of Fuganling granuleObjective On the basis of the function and treatment as well as the characters of the component of the herbs,we screened the technique designs,procedures and conditions to establish the quality standard of Fuganling granule.Methods The qualitative identification of the principal drugs(Huangxiangcaomuxi,Jigucao,Jinqiancao)in the formula was performed by TLC.Results In the chromatography of samples,the fluorescent speckles took an identical color at the corresponding position.Huangxiangcaomuxi,Jigucao,Jinqiancao could be detected by TLC,and the spot was clear,the degree of dissociation was good and easy to distinguish.Conclusions The established method is sensitive,rapid,accurate and well-reproducible and it can be used for the quality control of this product.Part 2 Experimental study on the effect of Fuganling granules in protecting immunity hepatic injuryObjective To study the effect of Fuganling(FGL)in protecting the mouse immunity hepatic injury which caused by Bacille calmette guerin(BCG)and Lipopolysaccharide (LPS),and observe the effect of FGL granule on mouse immumological function.Methods A total of 50 mice were injected visceral by BCG and LPS to establish immunological liver injury model,then were divided into 5 groups,4 groups were given 100mg/kg,50mg/kg,25mg/kg FGL and LianBenShuangZhi(80mg/kg) through tube feeding respectively,and the else 10 mice model were given normal saline.10 mice without injection were the normal control.Detecting the index of thymus,liver and spleen,detecting the activity of the alanine aminotransferase (ALT),aspartate aminotransferase(AST),MDA in serum,ultrastructure to verify the protect effect of FGL.Apply mouse hemolysin method to observe mouse immumological function.Results(1)Compare to the control group,the model group's thymus index decreased noticeably;liver index and spleen index increased significantly,The FGL in high-dosage,middle-dosage can increase the thymus index remarkably(P＜0.01).(2)The activity of ALT and AST in serum step up in a great scale(P＜0.01);In the difference,the other model group treated with FGL in high-dosage,middle-dosage or low-dosage can decrease the activity of ALT and AST.(3)FGL in difference dosage can decrease the content of MDA.(4)FGL in difference dosage can ameliorate mouse low-level hemolysin remarkably or very remarkably which caused by Cyclophosphamide.Conclusions This experiment construct the model of mouse immunity hepatic injury induced by BCG and LPS,FGL have a remarkably protective effect to this kind of immunity hepatic injury.FGL can improve the lower humoral immunity of mice caused by Cyclophosphamide,improve the ability of reticuloendothelial system of body to phagocytiae foreign body.
