ObjectiveTo investigate FOS,CHAT and VIP expression in the medullary visceral zone of the rats following cerebral hemorrhage(CH)complieated with multiple organ dysfunotion syndrome(MODS);TO discuss the potential role of vagus never and its transmittance CHAT,VIP in CMODS.Methods1.Model of Cerebral hemorrhage complicated with MODS was induced in rats by injecting CoilagenaseⅦinto the caudase putamen.Subdiaphragmatic vagotomy or vagus nerve stimulations by electricity was performed.2.90 Wister rats were randomly divided into five groups:normal control group(n=10),sham-operative group(n=10),subdiaphragrnatic vagotomy group (SDV,n-10),cerebral hemorrhage group(CH,n=20),cerebral hemorrhage+ subdiaphragmatic vagotomy group(CH+SDV,n=20),cerebral hemorrahge+vagus nerve stimulation group(CH+SIV,n=20).Incidences of systemic inflammatory response syndrome(SIRS)and MODS were diagnosed by their criteria.3.Changes of symptoms and signs including temperature,heart rate and respiration of each group were recored,the amount of WBC and function of liver and kidney were examined.Pathological changes of brain \lung \liver\intestines and kidney were recorded and pictures were taken through optical microscope.The area density of positive staining expressing FOS,CHAT and VIP in medullary visceral zone were analyzed for the relative content by immunohistochemistry(ABC). 4.Results were expressed as(?)+S,one-way-anova and T-test of SPSS 11.0 soft ware was used to evaluate the difference among groups.Results1.The rats of hemorrhage group had higher temperature and respiration than normal group,SDV group and sham-operative group(P<0.05),and the level of ALT,AST,BUN,Cr,LDH and LDH in CH group improved significantly(P<0.05).There were significant differences between CH+SDV group and normal control group, sham-operative group,SDV group,CH group(P<0.01)at above items.Another significant difference exists between CH+SIV group and CH+SDV group at above items(P<0.05).2.In CH group,CH+SDV group and CH+SIV group there were various inflammatory lesions on the organ tissue,in CH group the organ had significantly pathological changes at 24 hourpoint.In CH+SDV group the inflammatory reaction on lung and small intestine can be seen more serious than that in CH group.And the inflammation reaction was more serious in CH+SIV group than in CH group,while less serious than in CH+SDV group.In CH+SIV group,the peripheral organs had significantly pathological changes at 24 hour point,and the changes gradually recovered in 48 hours.3.The FOS or CHAT or VIP-positive neurons were observed in MVZ in CH group,CH+SDV group and CH+SIV group,in which positive neurons were denser than those in control group,sham-operative group(P<0.01).The expression of FOS,CHAT and VIP in CH+SDV group is less obvious than cerebral hemorrhage group. While the expression of FOS,CHAT and VIP in CH+SIV group is more obvious than CH group and CH+SDV group.ConclusionThe vagus nerve may play a positive role in cerebral hemorrhage followed MODS.Both CHAT and VIP may be the important transmittances of vagus nerve that affect the MVZ and peripheral organs in rats with CMODS.
