Background:Diabetes mellitus(DM)is an endocrine and metabolic disease caused by multiple etiological factors.Its characteristic is chronic hyperglycemia and meta bolic disorders of fats and proteins because of insulin secretion and /or efect d iscrepancy.Diabetes mellitus is a chronicity and lifetime disease which could c ause all kinds of diabetic complications.With the development of research in diabetic complications,hearing impairment caused people's notice increasingly. Hearing impairment in in type 2 diabetic patients happended unnoticedly which infects the life of patients deeply.There is no unified recognition about pathog enesy yet.Objective:The objective of the research was to study the effect of AGEs/RAGE in pathogencsis of diabetic hearing loss.Methods:All the maleWistar rats were mearsurcd auditory brainstem response(AB R).Then they were randomly divided into two groups.The rats of one group were subjected to 0.1%STZ throught the tail vein to induce diabetic model.T he other group was non-diabetic.After 24 weeks,all rats wcre measured auditor y brainstem rcsponse again then killed and blood was collectcd to measure fast ing plasma glucose(FPG),glycosylated hemoglobin(HbA1c)and AGEs.The RA GE was measured by immunohistochemistry.RT-PCR were performed to detcct the expression of RAGE mRNA in the cochlea tissue of all rats. Results:1.General data:In the course of experiment,the rats of DM group showed hyperdiuresis,polydipsia,polyphagia and emaciation markedly,furs were filthy reluster and growth slowly.The rats in NC group had no diabetic symptoms,their growth and nurtures were well.The body weight of the rats had no diference between two groups.After 24 weeks,the body weight of rats in DM was decreased obviously than that in NC group(P<0.001).FPG and HbA1c increased significantly in DM group than that in NC group(P<0.001).2.Plasma AGES:A fter 24 weeks,AGEs increased significantly in DM group than that in NC group.3.Before the experiment the ABR had no diference between 2 groups.24 weeks later the threshold and stage of latency of the ABRwere increased significantly in DM group than that in NC group.Hearing loss was remarkably associated with FBG,GHbA1c,LDL-c,AGEs and RAGE.4.The myocardial tissues of rat cochlea by HE showed stegnoticer lumens and thinner vascularis ductus cochlearis in DM group than NCgroup;the amount of spiral ganglion cell of DM group was smller than NCgroup.5.The RT-PCR showed that the gene expression of RAGE mRNA in DM group was more stronger than that in NC group.Conclusion:1.The type 2 diabetics rats had hearing loss,ABR can be used to evalue.2.Hearing loss was remarkably associated with FBG,GHbA1c LDL-c AGEs and RAGE.3.AGEs/RAGE participate the hearing loss of T2DM.
