Expression Of Bim MRNA In Leukemia And In Apoptosis The Relationship Between Ferritin And The Proapototic Protein Bim

Objective: Bim is a proapoptotic protein of the Bcl-2 family that shares only the short BH3 domain with other members. Human Bim gene located in the domain of chromosome of ten tumors including eight hematopoietic disease.The demonstration that Bim is essential for certain apoptotic responses and to prevent overproduction of hematopoietic cells suggests that it may be a tumor suppressor. Australia scientist Devireddy.L.R thinks that the machanism of apoptosis of Bim. Would related to Intracelular iron delivery and change the intracelucar iron delivery would be a effective way for treatment of leukemia. This experiment is to investigate the relationship between expression of Bim mRNA and leukemia ,and in apoptosis the relationship between ferritin and the proapototic protein Bim in leukemia patients,to reveal the machanism of apoptosis of Bim .Method:The expression of Bim mRNA in bone marrow mononuclear cell was detected by semi-quantaitaton RT-PCR technique in 30 leukemia patients and 10 controls.At the same time observe the ferritin in blood ,also by immunohistochemisty technique in 20 leukemia patients and 10 controls.Results:Bim mRNA was expressed in all group by statistical analysis ,the expression in leukemia patients was significantly lower than that in normol controls.(P<0.01).,but between the leukemia groups wasn't significantly diffferent. by immunohistochemisty the Bim expression in leukemia patients was lower than that in normol controls.(P<0.05). the ferritin in leukemia patients'blood was nagatively correlated with expression of Bim mRNA in bone marrow (r=--0.785
