Objective:To observe the effect of Yi Kun ning on function and structure in ovary of peri-menopause(PM) rats by the sex hormone regulation and ovarian repair of the natural aging rats.Methods:Thirty-two female wistar rats aged 11-13 months,which were identified to be PM,were divided into model group,Yi Kun ning group and western drug group. Additional 10 female rats aged 4 months were selected as the young control group. Using secretion levels of sex hormone on serum , vascular endothelial growth factor (VEGF),granulosa cell apoptosis and cell morphology,and other indicators to observe the effect of Yi kun ning on function and structure in ovary.Results:The results of serum sex hormone By RIA determination: the value of serum E2,P are higher,LH,FSH levels lower after treatment of Yi Kun ning and the western medicine group ( P<0.01 and P<0.05) ; The results of vascular endothelial growth factor by Immunohistochemical method:Yi Kun-treated group and control group of western medicine were in varying degrees positive,which were significantly different compared with the model group(P<0.01); The results of particles follicle apoptosis by in-situ end labeling (TUNEL):Although there were different levels of follicle particles apoptosis positive on Yi Kun-medicine group and the control group,but were significantly different compared with the model mice(P<0.01); The results of HE staining show Yi Kun-medicine group and the group of Western medicine were more follicles and less atretic follicles; As electron microscopy show nuclear membrane structure were clear, some particles nuclei and chromatin condense between Yi Kun-medicine group and western medicine group.Conclusions:This study confirmed that Chinese medicine Yi kun ning can increase serum E2 and P levels of PM rats,lower FSH and LH levels of serum; Play a role in promoting ovarian VEGF of PM rates; Play a inhibitory role on granulosa cell apoptosis,promopt that Yi Kun ning can prevent follicle atresia,thereby to get the mechanism of preventing and retarding aging of granulosa cell apoptosis. By changing the organizational structure and form of ovarian,it delays the aging role of ovarian.
