Objectives: To investigate the change of number and morphous of Langerhans cells(LCs) in the epiderm of seborrheic keratosis, keratoacanthoma, actinic keratosis, squamous cell carcinoma and basal cell carcinoma. And to discuss the relationship between the change of the number and morphous of LCs and sunlight-dependable skin tumors.Methods: Respectively, 15 paraffinum specimens of seborrheic keratosis, keratoacanthoma, actinic keratosis, squamous cell carcinoma and basal cell carcinoma were collected, 12 normal skin specimens of exposed position of adult normal were used for controlling. The expression of CD1a on the surface of LC of all specimens were detected by immunohistochemistry. Light microscope was used to observe the change of morphous of LCs, the average optical of unit area of each specimen was analysised by patho-image analysator.Results: The average optical(LC numbers) of unit area of normal skin was 1214.6±243.5, seborrheic keratosis was 1156.3±306.2, keratoacanthoma was 890.8±158.7, actinic keratosis was 305.1±124.2, basal cell carcinoma was 185.6±70.8, squamous cell carcinoma was 143.7±66.2. There was no significant difference in LC numbers between normal skin and seborrheic keratosis(P>0.05), and there was a statistically significant reduction in LC numbers for keratoacanthoma, actinic keratosis, squamous cell carcinoma and basal cell carcinoma compared with normal skin(P<0.05). There was no significant difference in LC numbers between seborrheic keratosis and keratoacanthoma, as well as between actinic keratosis and basal cell carcinoma, basal cell carcinoma and squamous cell carcinoma(P>0.05). But when seborrheic keratosis and keratoacanthoma were compared to actinic keratosis, squamous cell carcinoma and basal cell carcinoma respectively, actinic keratosis was compared to squamous cell carcinoma, there were significant difference(P<0.05). The morphous of LC in these skin tumors also changed significantly except for numbers, and was much more significant in malignant tumor of skin.Conclusions: Reduction in LC numbers and function maybe promote the development of skin tumor. The degree of the quantity and morphous change of LC could have a correlation with the degree of malignancy of skin tumors. Ultraviolet radiation maybe play an important role in the change of quantity and morphous of LC in these skin tumors.
