Objective: To approach the effects of mycobacterium infection in erythema nodosum, nodular vasculitis, erythema induratum, hypersensitivity angiitis. To explore the effective method to treat the above diseases.Methods: Paraffin-embedded skin tissues of erythema nodosum, nodular vasculitis, hypersensitivity angiitis, each were twenty; ten paraffin-embedded skin tissues of erythema induratum were enrolled. Among them male 33, female 37, age from 21-65, average 39.7, course of disease from 2 months to 2 years. All the paraffin-embedded skin tissues were from skin biopsy. The offered time was from to 2001-2007. The primer planning was: two pairs of primer were planned, the common primer of 16SrRNA of mycobacterium was 5'-AGAGTTTGATCCTGGCTCAG-3',5'-TGCACACAGGCCACA AGGGA-3',amplification product was 1030bp.The specific primer IS6110 of mycobacterium tuberculosis compound was 5'-CCTGCGAGCGTAGGCGTCGG-3',5'-CTCGTCCAGCGCCGCTTCGG-3', amplification product was 123bp.The primer was synthesized and purified by Saibaisheng Company. The positive control was Bacille Calmette Guerin (BCG).Twenty normal paraffin-embedded skin tissues were enrolled as negative control. DNA of mycobacterium from paraffin-embedded skin tissues were extracted, placed in PCR instrument and amplified. The amplficaion product was agarose gel electrophoretic, observed under the uviol lamp .Straps appeared in the place of 1030bp and 123bp were positive. Fisher's exact test was used in the results.Results: 1. 2 specimens were positive of 16SrRNA gene of Mycobacterium ,which were positive of DNA of Mycobacterium tuberculsis of 20 specimens of erythema nodosum .2. 2 specimens were positive of 16SrRNA gene of Mycobacterium ,none was positive of DNA of Mycobacterium tuberculsis of 20 specimens of nodular vasculitis.3. No specimen was positive of 16SrRNA gene of Mycobacterium,and none was positive of DNA of Mycobacterium tuberculsis of 10 specimens of erythema induratum . 4.No specimen was positive of 16SrRNA gene of Mycobacterium, and none was positive of DNA of Mycobacterium tuberculsis of 20 specimens of hypersensitivity angiitis. There was no significant deviation between the four diseases and Mycobacterium (P>0.05). There was no significant deviation between the four diseases and Mycobacterium tuberculosis(P>0.05).Conclusion: 1. The effects of mycobacterium infection were not relative to erythema nodosum, nodular vasculitis, erythema induratum, hypersensitivity angiitis. 2. The effects of mycobacterium tuberculosis infection were not relative to erythema nodosum, nodular vasculitis, erythema induratum, hypersensitivity angiitis.
