| Objective: To investigate the anti-tumor effect of self-made paclitaxel-carrying liposome microbubbles (PLM) radiated by fitted ultrasound on nude mice transplanted human ovarian carcinoma and its possible mechanism of action.Methods:We established nude mice transplanted SKOV3 human ovarian carcinoma.Then mice were randomly divided into 4 groups(5 mice in each group).Each was treated in a different way: PLM combined ultrasound group were treated with ultrasound fitted irradiation(output power 0.5W/cm2,output time 3min) soon after injecting PLM agent into tail vein(0.2ml);The mice of paclixtaxel chemotherapy group were treat by injecting chemotherapeutic drug paclixtaxel(0.2ml) into peritoncal;The PLM agent group were not treated with ultrasound probe irradiation after injecting PLM agent(0.2ml).The rate of tumor inhibition was calculated after sacrificing the animals at the end of experiment.Tumor tissues were obtained with the transmission electrion microscope(TEM). The basic fibroblast growth factor(bFGF),microvessel density(MVD)and nuclear associated antigen Ki-67 were measured immunohistochemically and analyzed through computer.Result: In the controls,the tumor volumes and weights inhibition rate was 0,the bFGF was 0.29248±0.01062,the MVD was 27.044±1.732,the Ki-67 was 0.30062±0.02032,whereas in the PLM combined ultrasound group,they were 55.41% and 52.77%,0.19049±0.01099,15.124±1.353,0.15699±0.01317 respectively,in the paclixtaxel chemotherapy group ,they were 36.48% and 28.06%,0.23304±0.01819,19.226±1.501,0.23271±0.01424 respectively,in PLM group they were 5.45% and 7.07%,0.27422±0.01342,25.228±1.151,0.28456±0.01588 respectively.Compared with control,the growth of tumor in PLM combined ultrasound group and paclixtaxel chemotherapy group was slow and the inhibitory effect of tumor was obvious(P<0.01). The Ki-67,MVD and bFGF expression all were decreased(P<0.01).But the forrer the inhibitory effect of tumor were much more obvious(p<0.01).Conclusion: Ultrasound fitted irradiation PLM agent has good therapeutic effect in the treatment on nude mice transplanted human ovarian carcinoma and makes it possible to target local release of drug .The mechanism of action may be due to its antiangiogenetic effect,interrupting blood-supply,inhibiting proliferation,inducing apoptosis. |
