| Countercurrent chromatography (CCC) is a liquid-liquid partition chromatography that no solid matrix is required, and can reach high efficient separation and recovery. Glucosinolates are anionic, hydrophilic plant secondary metabolites which are being recieved particular interest due to the role in anti-cancer and anti-bacterials of isothiocyanates from glucosinolates. Therefore, it was very important to separate and identify glucosinolates from cruciferous plants, which would be benefit to the development of products involving glucosinolates. In the present study, high-speed countercurrent chromatography (HSCCC) was applied to separate glucosinolates in the Cauliflower seed extract.A CCC solvent system composed of butanol-acetonitrile-10% ammiaonia sulfate (1: 0.5: 2.2) was found as an excellent solvent system to separate the glucosinolates in the crude extract using HSCCC-D1200 with a 1200 ml column, The glucosinolate components from CCC separations were purified by preparative HPLC to yield five glucosinolate monomers: 3-butenyl glucosinolate (â… ), 4-pentenyl glucosinolate (â…¡), 3-methylsulfinylpropyl glucosinolate (â…¢) , 4-methylsulfinylbutyl glucosinolate (â…£), 7-methylsulfmylheptyl glucosinolate (â…¤): The anti-allergic activity of seven compounds,4-pentenyl gluconsino -late,3-methylsulfinylpropyl gluconsinolate,4-methylsufinylbutyl glucosi -nolate, 7-methylsufinylheptyl gluconsinolate,allyl gluconsinolate,4-meth -ylsulphinyl-3- butenylglucosinolate was investigated by measuring their effects on hyaluronidase from bovine testis in vitro.The results showed that the activity strength wasâ…£>â…¦>â…¡>â…¤>â…¢>â… >â…¥at a dose of 1mg/ml. |
