Objective: The anticancer activity of an extract of Scurrula parasitic L. Parasitized on Nernium indicum Mill.(Nispex)on human Burkitt's lymphoma cells line CA46 and its molecular mechanism was studied here.Method: CA46 cells were exposed to Nispex at different dosages. Growth inhibition was detected by MTT assay and colony formation assay. Cell apoptosis was detected by staining of cells with AO-EB. TUNEL labeling method, DNA fragmentation and flow cytometry. The mRNA expression of bcl-2, bax, c-myc gene was detected by RT-PCR, nuclear transfer changes of NF-κB was detected and protein expression of Ikk, IκB, NF-κBp65, bax, bcl-2, caspase-3, PARP, c-myc related with cell apoptosis was detected by western blot.Result: Nispex remarkably inhibited the CA46 cells proliferation. Concomitantly, apoptosis was induced in CA46 cells, as measured by AO/EB fluorescence staining, TUNEL method, DNA fragmentation assay and AnnexinV-FITC/PI flow cytometry, CA46 cells apoptosis could be induced by Nispex. The bax gene mRNA on CA46 cells were increased and bcl-2, c-myc gene mRNA were decreased by Nispex treated, the content of NF-κB decreased obviouly and the Bax, Caspase-3 protein expression CA46 cells increased and I-κB, Ikk, NF-κBp65, Bcl-2, PARP, C-myc protein were decreased after Nispex treatment.Conclusion: Nispex could efficiently induce growth inhibition and apoptosis in CA46 cells, the inhibition of NF-κB signal pathway may be involved in CA46 cells apoptosis induced by Nispex.
