Objective:After establishing the animal model of periodontitis and chronic lung disease, Immunohistochemistry was used to evaluate the expression of TNF-αin the periodontal tissue and the lung tissue,in order to observe the relationship between them from the way of the cytokine.Methods:30 mice of Kunming specie,about 12 weeks old,were randomly divided into 2 group(group A and group B).Every group had 15 mice.A group was used the way of blowing gypsum for 7 weeks to establish the animal model of the chronic lung disease,groups A and B both ate the common forage during the 7 weeks.After establishing the model of the chronic lung disease,group A was divided into 3 subgroup to establish the model of the periodontitis by the way of the steel wire ligation and the high sugar diet.group A1 had the chronic lung disease and the normal periodontal tissue,group A2 had the chronic lung disease and the mild periodontitis,group A3 had the chronic lung disease and the heave periodontitis. group B used the same way as the group A to establish the the model of the periodontitis.group B1 was the contrast group.group B2 had the mild periodontitis. group B3 had the heave periodontitis.To contrast the depth of the periodontal pocket, the total alveolar bone loss,the proportion of the lymphocyte of the lung irrigation solution,pathological section of the periodontal tissue and the lung tissue.Immunohistochemistry were used to evaluate the expression of TNF-αin the periodontal tissue and the lung tissue,computer image analysis system were used for semi-quantitative analysis.Results:1.The periodontal depths of the mice show that group A3>group A2>group A1 (P<0.05);group B3>group B2>group B1(P<0.05),the total alveolar bone losses of the mice show that group A3>group A2>group A1(P<0.05);group B3>group B2> group B1(P<0.05),The compares between group A1 and group B1,group A2 and group B2,group A3 and group B3 show no differences in the periodontal depth and the total alveolar bone loss of the mice(P>0.05).2.the proportions of the lymphocyte of the lung irrigation solution show that group A3>group A2>group A1(P<0.05);group B3>group B2>group B1 (P<0.05).3.The pathological sections of the periodontal tissue show that:The periodontal fibers contact the cementum and the alveolar bone,and the cementum and the alveolar bone are complete in group A1 and group B1;In group A2,group B2, group A3 and group B3,the periodontal fibers contact the cementum and the alveolar bone,the blood vessels hyperemia in the periodontal membrane,the Howship lacunas appear in the alveolar bone,the osteoclast is in the Howship lacuna.The destruction of the periodontal tissue shows that the group A3 and B3 severer than the group A2 and B2.4.The pathological sections of the lung tissue show that:group A:The pulmonary alveolus aren't contact.The lung stromas thicken.The blood vessels dilate around the inflamed cells in the lung stroma.The severity of the inflammation shows group A3>group A2>group A1.group B:The inflamed cells are in the lung tissue,the pulmonary alveolus aren't contact.The severity of the inflammation show that group B3>group B2,and both of them are less severe than the group A.In the group B1,the lung tissue is normal,no inflamed cell.5.Immunohistochemistry show that:TNF-αmostly express among cytoplasm of periodontal ligament fibroblastes,osteoclasts,inflammation cell.semi-quantitative analysis results showed that:group A3>group A2>group A1(P<0.05);group B3>group B2>group B1(P<0.05).6.Immunohistochemistry show that:TNF-αmostly express among cytoplasm of lune cellula intersitialis,tracheal epithelium cell,inflammation cell and vaso-endothelial cell. semi-quantitative analysis results showed that:group A3>group A2>group A1 (P<0.05);group B3>group B2>group B1(P<0.05). Conclutions:1.Blowing gypsum for 7 weeks can establish the animal model of the chronic lung inflammation.Steel wire ligation and the high sugar diet for 3 weeks can stablish the animal model of the mild periodontitis.Steel wire ligation and the high sugar diet for 5 weeks can stablish the animal model of the mild periodontitis.The way of blowing gypsum plus steel wire ligation and the high sugar diet can stablish the animal model of the the chronic lung inflammation with periodontitis.2.TNF-αplay an important role on periodontitis,chronic lung inflammation and the relasionship between periodontitis and chronic lung inflammation3.The periodontitis can aggravate the inflammation of lung,and the more serious periodontitis the more serious lung disease.The periodontitis can lead to the lung disease,and the more serious periodontitis the more serious lung disease.The lung disease can't lead to and aggravate the periodontitis in the short time(3-5 weeks).
