Objectives To investigate effects of different doses of recombinant Insulin-like growth factorⅠ(rh IGF-Ⅰ)on Vascular Endothelial Growth Factor(VEGF)expresstion in MG-63.Methods Cell strain of MG-63 were cultured.The effects of rh IGF-Ⅰat different doses and different action times on VEGF expresstion patterns in MG-63 were observed with Reverse transcription polymerase chain reaction(RT-PCR).Results(1)In the present study,RT-PCR detected a different expression of VEGF mRNA andβ-action in the MG-63.(2)The levels of VEGF mRNA is increased obviously and signficantly at different doses of rh IGF-Ⅰ,about 2.2-fold,3.26-fold,4.37-fold,4.56-fold above the 0ng/ml,occoured at 24h.(3)The levels of VEGF mRNA in MG-63 is increased gradually as the increase of concentration in IGF-Ⅰfrom 0 to 50ng/ml.The levels of VEGF mRNA is increased signficantly at IGF-Ⅰat 0~50ng/ml compared with eath other(P<0.05). (4)The levels of VEGF mRNA is most increased and signficantly at 50ng/ml of IGF-Ⅰcompared with 0ng/ml(P<0.05).The levels of VEGF mRNA is not increased obviously and signficantly at 100ng/ml of IGF-Ⅰcompared with 50ng/ml(P>0.05).(5)It is statistically signficant(P<0.05),the levels of VEGF mRNA appeared to be increased obviously in rh IGF-Ⅰ-treated cultures(50ng/ml), showing peak induction at 24h.Conclusions(1)VEGF mRNA is expressed in the MG-63.(2)Rh IGF-Ⅰcan promote the expression of VEGF in dose-depentment and time-depentment manners.
