Diabetic Peripheral Neuropathic Pain(DPNP)is a common chronic complications of diabetes mellitus.It is also one of the most frequent reasons that causes acute and chronic pain in DM patients.The main clinical symptoms of DPNP are numbness of limbs,spontaneous pain,hyperalgesia,allodynia,even complying with anxiety and depression in Some severe patients.The pain degree is different,from slight pain to severe pain that can not be controlled by drugs and it extremely influences the patients both physiological function and quality of life.DPNP is a typical example of chronic neuralgia and the pathogenesis of it is very complicated.In the recent years,plenty researches have shown that some cells such as horizontal cell,gitter cell,macrophage,lymphocyte fibroblast et al,release a series of proinflammation cytokines,such as MIF,TNF-α,and IL-6.After nerve injury,these cytokines play an important role in mediating and maintaining neuralgia.However,it has not been reported domestic that whether these cytokines are different in the diabetic patients complying with pain or with no pain.Objective:comparing the content of cytokines (MIF,TNF-α,IL-6)in blood serum among the four groups:Normal control group,Diabetes mellitus group(DM),Diabetic Peripheral Neuropathic group(DPN),Diabetic Peripheral Neuropathic Pain group(DPNP),and investigating the role of these cytokines in DPNP.Methods:bolting qualified cases which were divided into four groups.Drawing 5ml blood in ulnar vein hollowly in the morning and put them into the test tubes containing no anticoagulant.Then,centrifuging the samples for 15 minutes(3000rpm,4℃)and taking blood serum as detecting materiae.These blood serum examples were preserved in refrigeraor with -70℃.We started to testify the blood serums after all the cases were collected.We detected these cytokines according to the kit descriptions and calculated the average absorbance values(A450)set of reference stands,control,samples.Drawing a standand curve,the OD value of standard example was veytical axis and its concentration was horizontal axis.we could get the examples' concentration by its OD values through standard curve.Results:The containing of MIF in the contyol group had significant difference in statistics compared with other three groups (p<0.001).There were significant statistics differences between the DM group and DPN group and DPNP group.(p<0.05).There were no significant statistics between DPN and DPNP.(p>0.05).The containing of TNF-αin the contyol group had significant difference in statistics compared with other three groups(p<0.001).There were significant statistics differences between the DM group and DPN group and DPNP group.(p<0.01).There were no significant statistics between DPN and DPNP.(p>0.05).The containing of IL-6 in the contyol group had significant difference in statistics compared with other three groups.(p<0.05).There were no significant statistics differences among the DM group,DPN group and DPNP group(p>0.05).there were no significant statistics between DPN and DPNP,either(p>0.05).Conclusions:1,cytokine MIF,TNF-α,IL-6 may play an important role in the pathogenesis of diabetes mellitus(DM)and Diabetic peripheral neuropathy(DPN);2,cytokine MIF,TNF-α,IL-6 do not play an important role in the development of Diabetic peripheral neuropathic pain(DPNP)。...
