[Objective] To investigate the effect of ADMA on endothelial cells and the therapeutic mechanism of Fenofibrate on atherosclerosis.[Methods] Cultured endothelial cells of human umbilical vein in vitro to detect the expression of MMP-9 and the production of NO after treated with different dosage of ADMA at certain time intervals by RT-PCR and Western blot, meanwhile, the interaction of Fenofibrate and ADMA.[Results] the expression of MMP-9 was up-regulated and inhibition of NO in the presence of ADMA, the effect of 10μmol/L ADMA group is the most notably, the difference is of significance. ADMA improved the expression level of MMP-9 and inhibited the synthesis of NO at different time points, and reached the peak at 48h. Meanwhile Fenofibrate reversed the effect of ADMA on the cells.[Conclusion] we found ADMA increased the expression of MMP-9 and decreased the production of NO ,and the impact of ADMA on the expression of MMP-9 and NO is both time and concentration dependent in the experimental models. Fenofibrate reversed the effect of ADMA on the expression of MMP-9 and promoted the synthesis of NO.
