Objective To explore the effects of recombinant human growth hormone (rhGH)on protein synthesis and structure of intestinal mucosa of trauma rats.Methods Following an adaptive feeding for one week,24 adult healthy Wistar rats received severe trauma and randomly divided into two groups by the method of completely randomized control.The control group(n=12)was treated with early enteral nutrition(EEN),which was direct feed of milk,and subcutaneously injected of normal saline.The experimental group(n=12)was received EEN combined with subcutaneously injected of rhGH.Four traumatic rats from each group were sacrificed at day 1,4 and 7 after trauma.The parameters of rats' weight,serum total protein,albumin,prealbumin were measured and analyzed.The morphology of ileum was observed and immunohistochemistry staining of interleukin 1(IL-1)on intestinal mucosa was performed.Results①Before the trauma,there was no significant difference between the experimental group and the control group on indexes of weight and serum proteins (P>0.05).②After the severe trauma,it showed no significant difference on the body weight of the two group on the 1st day(P>0.05).While on the 4th day,the body weight of experimental group began to be significant higher than that of the control group(P<0.05).This predominance still existed on the 7th day(P<0.05).③After the severe trauma,content of serum total protein of the two comparison groups showed no significant difference on all the term days(P>0.05).④There was also no significant difference on content of serum albumin on all the term days after the severe trauma(P>0.05).⑤After the severe trauma,content of prealbumin showed no significant difference(P>0.05)between the experimental group and the control group. While on the 4th day,content of prealbumin on the experimental group showed a significant priority(P<0.05).This priority also showed on the 7th day(P<0.05).⑥Observation on morphology of the intestinal mucosal after the severe trauma:both of groups put up the necrosis and exfoliation of intestinal mucosal villi from the end epithelial cells,obvious villi edema,and inflammatory cells infiltration on the 1st day. On the 4th day,both of the groups lightened on those symptoms which showed on the 1st day.But symptom of inflammatory cells infiltration on the experiment group was much lighter than the control group.And the villi was significant higher on the experiment group than the control ones.On the 7th day,the height of villi slightly enhanced,but inflammatory cells infiltration was sill be observed in the control group. But in the experimental group an integrated structure of intestinal mucosal villi and no edema showed.⑦IL-1 in the intestinal mucosal tissue:the level of IL-1 increased in both of groups.This was most obvious on the 1st injure day.As time went on,it had a gradually decreasing.Comparing with the two groups,IL-1 level showed no significant difference(P>0.05)on the 1st day.On the 4th day,with a wholly decrease, level of IL-1 showed a significant decline in the Experimental group comparing with the control one(P<0.05).On the 7th day,along with a further wholly decreasing,the decline in the experimental group was further accelerated,its IL-1 level was significant lower than the control group(P<0.05).Conclusion An EEN combined with rhGH treatment is superior single EEN on severe trauma rats in promoting protein synthesis,maintaining intact structure of intestinal mucosa,promoting villi growth and reducing inflammatory response.
