| Objective To investigate the protective effects and possible mechanisms on oxidative damage in the lens.Methods Fifty femal wistar rats were randomly divided into five groups:normal(A),model(B),ovariectomy(C),ovariectomy with estrodiol replacement therapy(D),sham operated(E).After four weeks of opration,D group were given three daily injections of 0.5ug 17—β-estradiol,dissolved in benzyl alcohol(1:3,vol/vol).C group received vehicle alone.Making Uv damage model of anterior segment of eyes except A.24 hour later,rats were killed before removal of eyes.Lenses were carefully dissected free from surrounding ocular tissues.Take lens capsule to measure the contents of malondialdehyde,the activity of superoxide dismutase.Take previous lens capsule to measure the apoptosis rate of lens epithelial cells.The super microstructure of apoptotic cells were observed with TEM.Results 1.It showed serious peroxidation damage in ovariectomy group compared with other groups,but expressions in B,C and E were similar.2.In examination of lens antioxidative defense system,the level of MDA in ovariectomy group increased as compared with other groups(p<0.05),while the levels of SOD, were lower than other groups(p<0.05),there were no significiant differences among model,ovariectomy with estrodiol replacement therapy,sham operated.3.The apoptosis rate of normal group was lower than the others(p<0.05).The model,ovariectomy with estrodiol replacement and sham operated were lower than ovariectomy(p<0.05)and there were no significiant differences among the first three. 4.In addition to the A,have both been typical characteristic changes of apoptosis by TEM.Conclusions Uv induced oxidative damage in lens protein and epithelial cells to apoptosis,resulting in the formation of cataracts.Estrogen as an anti-oxidant protection,may become for prevention and treatment of drug cataract provide experimental basis. |
