The Effects Of 2-(2-(m-tolyloxymethyl)-1H-benzo[d] Midazol-1-yl) Acetic Acid And 2-(2-(p-tolyloxymethyl) 1H-benzo[d] Midazol-1-yl) Acetic Acid Antifungal

Objective Development of new antifungal through in vitro and animal experiments.Methods and materials(1)Antibacterial test in vitro Using constants broth dilution method.The experiment is divided into test group(Groupâ… ),blank control group (Groupâ…¡)and drugs(fluconazole)control group(Groupâ…¢).Groupâ… and groupâ…¡is divided into four subgroups,each subgroup has 10 test-tube,the three previous subgroups by adding MH broth,the last subgroup by adding RPMI 1640 medium.Groupâ…¢only has one subgroup,by adding RPMI 1640 medium.Staphylococcus aureus ATCC25923, Pseudomonas aeruginosa ATCC27853,E.coli ATCC25922 be added into MH broth each, after incubating for 24 hours,and Candida albicans ATCC90028 incubating for 48 hours,then we observe the test results.(2)Animal experiments Using Wistar rats as experimental animals,experimental animal were randomly divided into three groups,each group has 10 rats. Treatment groups include of two group,2-(2-(m-tolyloxymethyl)-1H-benzo[d]midazol-1-yl) acetic acid is named of groupâ… ,and 2-(2-(p-tolyloxymethyl)-1H-benzo[d]midazol-1-yl) acetic acid is named of groupâ…¡,the last group is the control group,groupâ…¢.Test induced firstly by cyclophosphamide,producing hemorrhagic cystitis,Then introduce Candida albicans into bladder,make the inflammation aggravation.Removal of the rat's bladder after 24 hours,then make pathological section.Results(1)Antibacterial test in vitro Comound 2-(2-(m-tolyloxymethyl) -1H-benzo[d]midazol-1-yl)acetic acid(sha2)MIC for Candida albicans ATCC90028 is of 7.81ug/ml;Comound 2-(2-(p-tolyloxymethyl)-1H-benzo[d]midazol-l-yl)acetic acid(sha₃) MIC for Candida albicans ATCC90028 is of 1.95ug/ml;Fluconazole injection MIC for Candida albicans ATCC90028 is of 0.625ug/ml.(2)Animal test results Bladder wall sections were observed under microscope,Group.â…¢could be see obvious mucosal injury,It's succession is damaged,Submucosal exfoliated,Bladder wall tissue could be see extensive bleeding and congestive phenomenon,groupâ… andâ…¡similar to the groupâ…¢that has the same pathological features,Mucosa fracture,exfoliated,Some parts of muscular layer have bleeding and congestive phenomenon,But these phenomenon reduced significantly contrase to groupâ…¢.Conclusion The test results,showed that in vitro test and animal experiment, compound 2-(2-(m-tolyloxymethyl)-1H-benzo[d]midazol-l-yl)acetic acid(sha₂)and 2-(2-(p-tolyloxymethyl)-1H-benzo[d]midazol-1-yl)acetic acid(sha₃)have antifungal effect,and provided preliminary work for clinical that searching for a new antifungal.
