Objectives To approach the mechanism of action of electro-acupuncture (EA)about improving learning and memory ability in vascular dementia by studying the changes of expression of synaptophysin and NMDAR-2B on hippocampal formation of vascular dementia rats in the chronic cerebral ischemia after electro-acupuncture's treatment.Methods SD rats with normal learning and memory were divided into blank control group(A group),sham operation group(B group),operation group by random after they were tested by Morris Water Maze.Permanent bilateral occlusion of the common carotid arteries were used to established the vascular dementia models in operation group rats.After six weeks operation group rats were tested by Morris Water Maze again,then the rats with degraded ability of learning and memory were divided into as vascular dementia group(C group),electro-acupuncture(EA)nonacupoint group(D group)and electro-acupuncture(EA)acupoint group(E group). "Baihui","dazhui" and bilateral "shenshu" were selected as electro-acupuncture acupoint in E group rats,bilateral part away from spine(nonacupoint)were acupunctured in D group,the two group rats were stimulated with EA of an equal condition for 30 days,meanwhile A group,B group and C group were fed normally without any treatment.After eleven weeks(EA incentive were finished)learning and memory ability of five groups rats were detected by Morris Water Maze again.Subsequently, consecutive corona-frozen section of brain tissues from chiasm opticum to bulla fornicis were used to assayed.Immunohistochemistry were applied to detected locatively and semiquantitatively the changes of synaptophysin and NMDA-R proteins,and hybridization in situ were applied to detected locatively and semiquantitatively the changes of synaptophysin and NMDA-R mRNAs.Resultsâ‘ Six weeks after operation learning and memory ability of C,D and E groups rats were lower than A and B groups,and learning and memory ability of E group rats were improved by EA,but were not restored until the eleventh week after operation.â‘¡The expression of synaptophysin and NMDAR-2B proteins and mRNAs of C group rats were significantly lower than those of A group rats,and the expression of synaptophysin and NMDAR-2B proteins and mRNAs of the E group rats were significantly higher than those of C groupratsinhippocampus CA1 region and dentate gyms.The expression of both synaptophysin protein and mRNA and NMDAR-2B protein and mRNA in every group rats were positive correlation.â‘¢Protein and mRNA of synaptophysin in every group rats were positive correlation to those of NMDAR-2B in hippocampus CAt region and dentate gyrus.Conclusionsâ‘ The reliable vascular dementia rat model could be established by permanent bilateral occlusion of the common carotid arteries.â‘¡At the same time expression changes of synaptophysin and NMDAR-2B participated the development of vascular dementia induced by the chronic cerebral ischemia.â‘¢Electro-acupuncture in "Baihui","dazbui",bilateral "shenshu" could simultaneously improve expression of proteins and mRNAs of synaptophysin and NMDAR-2B,thus the learning and memory ability was amended.
