Objective:Taxinol(TAX)resistant gastric cancer cell line SGC7901/TAX was established in vitro,observating its biological characteristics , detecting the expression of its mdr1 ,approaching initially the occurance of multidrug resistance.Methods:TAX-resistant gastric cancer cell line SGC7901/TAX was obtained by culture of gastric cancer cell line SGC7901 in vitro with intermittently increasing the concentration of TAX in the culture medium for about seven months.The appearance of the SGC7901 and SGC7901/TAX was observated in light microscope after HE staining, the sensibility to chemotherapeutic drug of the two cell lines was compared through MTT experiment , the expression of their mdr1 was detected by RT-PCR.Results: After inducing with TAX for about seven months, SGC7901/TAX was obtained.Comparing with SGC7901 , SGC7901/TAX was not coincident,some of them were spindle,large cells appeared;MTT experiment displayed that SGC7901/TAX developed a cross resistance to TAX,5-Fu and CDDP,IC50 as follow: (1),TAX:a,SGC7901:27.09±0.12μg/L,b,SGC7901/TAX:135.50±0.51μg/L,the difference had statistical significance(P<0.001);(2),5-Fu:a,SGC7901:106.87±0.48μg/L,b,SGC7901/TAX:319.40±0.98μg/L , the difference had statistical significance (P<0.001);(3),CDDP:a,SGC7901:317.85±1.33μg/L,b,SGC7901/TAX:519.45±5.73μg/L,the difference had statistical significance(P<0.001)。The drug resistance indexs of cell line SGC7901/TAX to TAX,5-Fu and CDDP were 5.00,2.99and 1.63.The expression level of mdr1mRNA was detected by RT-PCR,that in SGC7901 was 0.3065±0.0374,that in SGC7901/TAX was 0.8954±0.0323,the difference had statistical significance(P<0.001).Conclusions:1. SGC7901/TAX can be established by culture of gastric cancer cell line SGC7901 in vitro with intermittently increasing the concentration of TAX in the culture medium;2. The occurance of multidrug resistance of SGC7901/TAX had relationship to the expression of mdr1;3. SGC7901/TAX developed a cross resistance to TAX,5-Fu and CDDP.
