Objective: To investigate the results of effect and mechanism of tamoxifen(TAM) on the growth inhibition and apoptosis inducement in prostate cancer cell PC-3, which may provide a new way for the therapy of prostate cancer.Methods: Cultures of prostate cancer cell were exposured to tamoxifen at the different concentrations of 0 mol·L-1,10-6mol·L-1,10-5mol·L-1,10-4mol·L-1 and 10-3mol·L-1 respectively.The effects of tamoxifen on growth inhibition and apoptosis inducement were determinated with morphometry,MTT method and Tunnel.The activity of caspase-3 were determinated with flow cytometry.Rusults: The hyperplasy of PC-3 were significantly inhibited in the 10-5mol·L-1,10-4mol·L-1 and 10-3mol·L-1 groups(P<0.05), but the effcects were nonsignificant difference in 0 mol·L-1 and 10-6mol·L-1 groups.The activity of caspase-3 in 10-4mol·L-1 and 10-3mol·L-1groups were significantly higher than other groups(P<0.05),but the expression of active caspase-3 was nonsignificant difference in 0 mol·L-1,10-6mol·L-1 and 10-5mol·L-1 groups. The expression of active caspase-3 were not differen in the time.Conclusions: The growth inhibition and apoptosis inducement could be taken by tamoxifen in prostate cancer cell PC-3 in a dose and time-dependent manner.The mechanism was that the high activity of caspase-3 was caused by tamoxifen,which led to the apoptosis of PC-3.
