[OBJECTIVE]:To investigate the effect of CD437 on apoptosis of human hepatoblastoma cell HepG2 and the possibility role of the alteration of mitochondrial membrane potentials(MMP) and ROS.[METHODS] : The effect of CD437 on the proliferation of Hepatoblastoma cells was measured with colorimetry. Annexin V-FITC/PI double fluorescent labeling was used to display the cell apoptosis. The mitochondrial with Rhodamine 123 probes staining were detected by fluorescent microscopy and the changes of MMP were analyzed by flow cytometry.The ROS with DCFH-DA probes staining were detected by fluorescent microscopy and the changes of ROS were analyzed by flow cytometry.[RESULTS] : CD437 significantly inhibited the growth of HepG2 cells.ROS rised and MMP fell while CD437 induced the apoptosis.[CONCLUSION]:The rose of ROS and disruption of the MMP may be one of the mechanisms that CD437 induces apoptosis of HepG2 cells.
