Object To investigate the effects of Bone marrow-derived mesenchymal stem cells transplanting on recovery of neurological functions in focal cerebral infarction in rats and its molecular mechanism. Methods Bone marrow-derived mesenchymal stem cells were harvested and purified following a previously described adherence technique. Rats permenant focal cerebral ischemia model was established with modified Longa's method. BMSCs were labeled by Dapi before the transplantation. One day after right middle cerebral artery occlusion(MCAO),1×106 cells were injected into the lateral ventricle of BMSCs-treated group rats. Before sacrificed and at 4d,7d and 14d after MCAO,the neurological functions were tested by balance beam,rota-rod and screen prehensile. The IL-10 was detected by reverse transcriptase-polymerase chain reaction(RT-PCR) and ELISA , the survivin and Synaptophysin were measured by RT-PCR and immunohistochemical method at 4d,7d and 14d after MCAO. Results Dapi staining positive cells were observed around the cerebral infarcted area in the BMSCs-treated group. By compared with the MCAO group and the vehicle group,the neurological functions were better in BMSCs-treated group at 7d,14d after MCAO (P<0.05 or P<0.01),and IL-10,survivin and synaptophysin around the cerebral infarcted area were significantly upregulated at 4d,7d,14d after MCAO too (all P<0.01). There was no significant differences of neurological functions in rats'brains 3d post MCAO among BMSCs-treated group,MCAO group and the vehicle group.Conclusions:1,BMSCs Transplanting can improve the neurological functions,and its Mechanism was related to upregulate IL-10 expression expression after MCAO in rats.2,BMSCs Transplanting can improve the neurological functions by upregulating survivin expression after MCAO in rats.3,BMSCs Transplanting could promote the recovery of neurological functions and synaptophysin synthesis in the brains in MCAO rats,which suggested that BMSCs Transplanting play a important role in the recovery of neurological functions post cerebral ischemia.
