Object Established the SD rat mode of benign prostatic hyperplasia (BPH), investigate the effcts of Finasteride on Vascular Endothelial Growth Factor (VEGF) and microvessel density (MVD) in the rat prostate tissue .Methods 56 male SD rats,weight:300±25g.Randomly selected 8 to feed normally , as a contorl group benign prostatic hyperplasia (I);The remaining 40 subcutaneous injection of testosterone propionate in neck every other day (For 28 days) to establish the rat mode of BPH. Randomly execute 8 rats (H) and get the prostate to do HE stain and divided the remaining rat mode of BPH (40)into 5 groups in the 29 days: A group lavage to thenomal saline(15ml/kg ), B﹑C﹑D﹑E group lavage to Finasteride of solution(1.0mg/kg), the duration of one week, respectively, two weeks, three weeks, four weeks. All rats feed one week normally after gavage finished of the E group,then executed them .Removaled and weighed the prostae ,measured the volume using the way of replace water.Observed the pathomorphology by light microscope and assessed VEGF and CD31 expression in the rat by immuno-histochemical method.As to anti-CD31 antibody detection of vascular endothelial cells,thus calculated MVD.Result①Compared with the I control group ,the benign prostatic hypertrophy model group weight of prostate added obviously,p<0.001(α=0.05), the difference was statistical significance.②Compared with control group ,the prostate in groups of the rat mode of BPH align compact,the lumen of gland change great and partial of glandular organ expand,glandular epithelium hyperplasia obviously,the layer of partial increased and align chaotic,the most glandular epithelium extend into intracavitary as corpora mammillaria .The prostate has been proved hyperplasia of prostate gland by HE stain. Established the rat mode of BPH has succeed.③The expressions of VEGF and MVD in A group were significantly higher than those in B,C,D and E groups,the difference was statistical significance (P<0.05); The expressions of VEGF and MVD in B group were significantly higher than those in D and E groups,the difference was statistical significance (P<0.05); The expressions of VEGF and MVD in C were significantly higher than those in D and E groups;the difference between B and C,D and E was not statistical significance (P>0.05).④The expression of VEGF and CD31 positive correlation, (C =0.865, P<0.001)。Conclusion Finasteride could effectly inhibit the expression of VEGF in the rat prostate tissue and degrade the MVD, and the degree of inhibition is concerned with the action time of Finasteride.
