| Objective:To research the effect of the calcineurin (CaN) on the cardiacmyocyte apoptosis induced by ischemia reperfusion and amlodipine.Methods:The left anterior descending artery in Wistar rat was ligated to establish ischemia reperfusion model (3 hours reperfusion after 30 minutes ischemia). 36 rats were divided into six groups based upon whether amlodipine intervention was used (6 /each group): sham-operated A and B group,ischemia/reperfusion A and B group(I/R), ischemia/reperfusion +amlodipine pretreatment group , ischemia/reperfusion + amlodipine treatment group(amlodipine was administered at a daily dose of 10mg.kg-1·d-1 for 7 days). The apoptosis rate of cardiacmyocyte was tested through flow cytometry and the levels of calcineurin were tested through RT-PCR.Results:1.CaNmRNA changes in the expression level of comparison: I / R A and B group CaNmRNA expression was higher than that in sham-operated A and B group, a statistically significant difference (P <0.05).Amlodipine pretreatment group and treatment group CaNmRNA myocardial cells than the expression level of I/R group was significantly decreased (P <0.05).2.Comparison of changes in the rate of apoptosis: I / R A and B group apoptosis was higher than that of sham-operated A and B group, a statistically significant difference (P <0.05). Amlodipine pretreatment group and treatment group cardiomyocyte apoptosis rate than I / R group was significantly decreased(P <0.05).Conclusions: CaN was suggested to own positive effect on the cardiac myocytes apoptosis during I/R. The both intervention of amlodipine pretreatment and therapy was helpful on I/R myocardium, in which amlodipine diminished CaN activity and the cardiomyocyte apoptosis.
