Objective:Sporotrichosis is a skin disease of deep fungal disease in a more common, the pathogenic bacteria is sporothrix schenckii, skin infections can also be fixed in disseminated systemic, immunocompromised patients, such as AIDS patients. The infection can spread to internal organs often, and even life-threatening Sporothrix schenckii major encroach the skin and easily injured more common in exposed positions, mainly divided into the skin type and skin appearance. The former can be divided into fixed cutaneous sprortrichosis, regional lymphangitic sporotrichosis, skin disseminated type. Different clinical manifestations. is of Sporothrix of duplex pathogenic fungi, which is at room temperature of the mycelium, the body of 37℃for the yeast, the yeast phase is the disease. for the two-phase fungi and diverse clinical manifestations, to the Sporothrix of the mechanism of disease has caused great difficulties to the pathogenic mechanism is not clear so far, in research and exploration stage. This study by the different different types of Sporothrix to DNA extraction and random amplified polymorphic DNA (Random Amplified Polymorphic DNA, RAPD) analysis, through animal experiments and indirect immunofluorescence to observe different clinical types of Sporothrix with the ability of extracellular matrix proteins different from the molecular level to explore different clinical types of Sporothrix pathogenic strain of the differences, in order to explore the pathogenic mechanism and provide a theoretical basis.Methods:1.Collection of different clinical types of Sporothrix isolates and genomic DNA of s.schenckii isolates were amplified by random amplified polymorphic DNA (RAPD) analysis.2.Suspensions of different isolates of s.schenckii were inoculated into BALB / c mice in order to observe their pathogenicity 3.Histopathology of the skin was studied to determine the numbers and distribution of spores.4.Indirect immuno- fluorescence assay to explore extracellular matrix protein fibronectin binding of sporotrix schenckii.Results:1.The electrophoretic profiles of PCR products differed in the isolates from different clinical patterns of sporotrichosis:1400 bp \ 800bp \ 700bp \ 500bp in the regional lymphangitic sporotrichosis, 2500 bp \ 1400bp \ 1000bp \ 700bp in fixed cutaneous sprortrichosis.2.The earlier onset of illness,wider distribution of lesions were observed in the BALB/c mice inoculated with isolates from regional lymphangitic sporotrichosis than fixed cutaneous sprortrichosis.3.Histopathologically :more spores were found in the mice inoculated by isolates from regional lymphangitic sporotrichosis than fixed cutaneous sporotrichosis.4.Indirect immunofluorescence results showed that different clinical patterns of Sporothrichosis's has different ability to bind extracellular matrix protein fibronectin.Regional lymphangitic sporotrichosis showed intensity fluorescence,but the yeast cell of fixed cutaneous sprortrichosis presented a faint fluorescence. Regional lymphangitic sporotrichosis has the more strong combination than fixed cutaneous sprortrichosis.Controls performed using a rabbit normal serum instead of the primary antibody were negative,attesting the specificity of the labeling.Conclusions:1.Different clinical patterns of Sporothrichosis has different virulence.2.After the Sporothrichosis cell culture , extract DNA and random amplified polymorphic DNA (RAPD) analysis.The electrophoretic profiles of PCR products differed in the isolates from different clinical patterns of sporotrichosis.3.The earlier onset of illness,wider distribution of lesions were observed in the BALB/c mice inoculated with isolates from regional lymphangitic sporotrichosis than fixed cutaneous sprortrichosis.4.Indirect immunofluorescence results showed that different clinical patterns of Sporothrichosis's has different ability to bind extracellular matrix protein fibronectin .Regional lymphangitic sporotrichosis has the more strong combination than fixed cutaneous sprortrichosis.Through this experiment, suggesting that different clinical patterns of Sporothrix has its pathogenicity different, Sporothrichosis pathogenic mechanism needs further study.
