Background:It's well-known that dyslipidemia is important,Its mechanism has not yet been clarified. With the discovery of peroxiisome proliferator-activated receptors , the association of peroxiisome proliferator-activated receptor alpha(PPARα) with dyslipidemia is being received a lot of attention. Recent studies have shown that PPARαis widely expressed in all organs and tissue,such as liver, kidney, heart, small intestine and brown adipose tissue. PPARαregulates the transcription of key proteins involved in lipid metabolism, immunity reaction, cell differentiation ,as well as inflammatory reactions.It's shown recently that C2528 is associated with an earlier mean age of diagnosis with type 2 diabetes and dyslipidemia. Therefore, we study PPARαgene C2528G polyphism and explore the association of peroxiisome proliferator-activated receptor alpha(PPARα) gene C2528G variation with dyslipidemia in Dalian of China.Objective:The purpose of this study is to explore the correlation of PPARαgene G2538 polyphism to dyslipidemia.Methods:Dylipidemia group subjects (one of TC and TG is higher than normal at least) are composed of 100 patients(58 men and 42 women),whose average age is 53.48±13.14 year, and all of them is Han people of Dalian.Normal control group subjects are composed of 100 healthy people (48 men and 52 women),all of them are non-diabetes,non ischaemic-heart-disease people,and with the normal blood pressure and lipid indicators,and the average age is 51.57±11.58 year,.All the subjects selected are excepted for such problems as tumor, sevier liver and renal dysfunction,cardia insufficiency and the prensence of stress factors. All of them is Han people in Dalian .Extracted peripheral blood white blood cell genomic DNA to detect PPARαG2528 polymorphism by the application of polymerase chain reaction-restriction fragment length polymorphism (PCR-PFLP), and calculate genetype and allele frequency of each group,as well as differences; simultaneously compare with body mass index (BMI), triglyceride (TG), total cholesterol (TC), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), blood urea nitrogen (BUN), camine (CR), uric acid (UA), fasting blood glucose (FBG), glutamic pyruvic transaminase (GPT), glutamic oxalacetic transaminase (GOT) between the two groups.Results:The overall frequencies of CC, GC, GG in normal control group and in dylipidemia group were 0, 11%, 89%, and 6%, 29%, and 65% respectively. There was statistical difference in the genetype frequency distribution between the two groups by check-up (p<0.01). The C2528 allele frequency is 5.5%, and G2528 allele frequency is 94.5% in normal control group; and the C2528 allele frequency is 20.5%, and G2528 allele frequency is 79.5% in dyslipidemia group. There is statistical difference between them by check-up (p<0.01). In normal control group G2528 allele frequency is higher than dyslipidemia group.Conclusion:The association of PPARαC2528G gene with dyslipidemia is evident in Chinese of Dalian. In the normal control group G2528 sllele frequency is more elevatory than the dyslipidemia group.
