Objective Very serious epidemic of tuberculosis in China. Retreatment of patients is a large number. Retreatment of pulmonary tuberculosis is a major problem in the control of TB. A study showed that susceptibility genes associated with the retreatment of pulmonary tuberculosis. To investigate the distribution of the vitamin D receptor (VDR) gene polymorphisms of Chinese Han population among new and retreated pulmonary tuberculosis. For further study on relation between VDR gene polymorphisms and new and recurred pulmonary tuberculosis.Method Hospital-based case-control study design was used. We selected 30 new pulmo nary tuberculosis and 30 recurred patients and 30 ethnically matched healthy controls. Whole blood extraction of genomic DNA, amplified by PCR.We typed the polymorphisms of VDR gene, Taq I and Fok I loci, with SNaPshot SNP genotyping techniques.Result①Three groups in the age difference not statistically significant;②Genotype of VDR-ff were significantly associated with susceptibility to tuberculosis, the OR (95 % CI ) was 6.53(1.26~33.9)when comparing with FF genotype, P=0.03. The other genotypes (FokI sites Ff, ff genotype; Taq I TT, Tt, tt genotype) in the distribution among the three groups was no significant difference(P>0.05);③No significant difference was observed on the distribution of TaqI and FokI polymorphisms between new and recurred patients with pulmonary tuberculosis(P>0.05), Joint analysis showed that the genotypes in each group there was no significant difference..Conclusion①The VDR-ff genotype were associated with susceptibility to pulmonary tuberculosis in Chinese Han population, ff genotype in the TB patients probability is 6.53 times than in the health.②Further study was required on weather the TaqI and FokI polymorphisms loci is existed differences in the distribution. VDR gene polymorphism on recurred PTB may be the same with the new PTB.③With the further understanding of VDR gene polymorphism, To explain the molecular mechanism the body's immune system to Mycobacterium tuberculosis infection and the molecular mechanism of recurred PTB;and to improve the prevention and control of tuberculosis.
